水稻細(xì)胞質(zhì)雄性不育恢復(fù)基因Rf6功能標(biāo)記開發(fā)及微核心種質(zhì)中Rf6基因的篩選

【摘要】:核恢復(fù)基因Rf6是紅蓮型雜交水稻特有的新恢復(fù)基因,為了尋找更多具有Rf6恢復(fù)基因的種質(zhì),以擴(kuò)展紅蓮型雜交水稻的恢復(fù)系來源,組配更多優(yōu)良雜交品系,利用Rf6及其等位基因的序列差異,開發(fā)Rf6基因的功能標(biāo)記,并利用該標(biāo)記對(duì)中國水稻微核心種質(zhì)進(jìn)行PCR擴(kuò)增,篩選具有Rf6恢復(fù)基因的種質(zhì)。結(jié)果表明,在324 bp插入片段(Rf6較其等位基因插入324 bp)兩側(cè)設(shè)計(jì)引物(正向引物:5'-ATGACAAGAGGACCAGCGATGCAATGG-3',反向引物:5'-ATTCTTGCAGAGATAGACCATGAGCGTG-3'),利用該引物可擴(kuò)增出條帶清晰且無雜帶的差異片段,可用于Rf6基因的鑒定。利用該標(biāo)記在197份中國水稻微核心種質(zhì)中篩選出22份具有Rf6恢復(fù)基因的種質(zhì),可為拓寬紅蓮型雜交水稻的恢復(fù)系來源提供材料。
[Abstract]:The nuclear restorer gene Rf6 is a unique new restorer gene in red lotus hybrid rice.
