FoxO1及自噬相關(guān)基因Beclin1和MAPLC3在胃癌組織的表達(dá)及意義

【摘要】:目的探討叉頭轉(zhuǎn)錄因子O亞家族1(FoxO1)及自噬相關(guān)基因Beclin1和微管相關(guān)蛋白輕鏈3(MAPLC3)在胃癌組織的表達(dá)及意義。方法收集本院2015年1月至2016年6月收治的82例手術(shù)切除的胃癌組織(胃癌組)及67例癌旁正常組織(對照組),采用熒光實(shí)時定量PCR(qPCR)法檢測FoxO1、Beclin1及MAPLC3水平,比較組間的水平變化并分析胃癌組中FoxO1、Beclin1及MAPLC3表達(dá)變化的相關(guān)性,分析組織FoxO1、Beclin1及MAPLC3水平變化與胃癌臨床病理參數(shù)[性別、年齡、TNM分期、分化程度、部位、淋巴結(jié)轉(zhuǎn)移及癌胚抗原(CEA)水平]的關(guān)系。結(jié)果qPCR檢測結(jié)果顯示,胃癌組的FoxO1、Beclin1及MAPLC3水平依次為(0.489±0.032)、(0.148±0.012)和(0.581±0.038),均低于對照組,差異有統(tǒng)計學(xué)意義(P0.05);胃癌組的FoxO1水平與Beclin1水平呈正相關(guān)(r=0.270,P=0.014),Beclin1水平與MAPLC3水平呈正相關(guān)(r=0.351,P=0.001),但FoxO1水平與MAPLC3水平無關(guān)(r=0.208,P=0.061);FoxO1表達(dá)與TNM分期、分化程度及淋巴結(jié)轉(zhuǎn)移均有關(guān),Beclin1表達(dá)與TNM分期、淋巴結(jié)轉(zhuǎn)移及CEA水平均有關(guān),且MAPLC3表達(dá)與TNM分期、分化程度及CEA水平均有關(guān)。結(jié)論FoxO1、Beclin1及MAPLC3在胃癌組織中表達(dá)降低,且均與TNM分期有關(guān),對胃癌診斷及病情評估有一定價值。
[Abstract]:Objective to investigate the expression and significance of fork transcription factor O subfamily 1 (FoxO1), autophagy related gene Beclin1 and microtubule associated protein light chain 3 (MAPLC3) in gastric cancer. Methods from January 2015 to June 2016, 82 cases of gastric cancer (gastric cancer group) and 67 cases of paracancerous normal tissues (control group) were collected. The levels of FoxO1,Beclin1 and MAPLC3 were detected by fluorescence real-time quantitative PCR (qPCR). The changes of FoxO1,Beclin1 and MAPLC3 expression in gastric cancer group were compared, and the changes of FoxO1,Beclin1 and MAPLC3 levels in gastric cancer group were analyzed.
