基于生物信息學(xué)方法的蕎麥屬microRNAs及其靶基因預(yù)測

【摘要】:MicroRNA是一類具有轉(zhuǎn)錄后基因調(diào)控功能的非編碼小分子RNA。以傳統(tǒng)的試驗(yàn)方法發(fā)現(xiàn)和鑒定新的miRNA是一項(xiàng)繁瑣的工作。以生物信息學(xué)方法從數(shù)據(jù)庫中篩選miRNAs及其靶基因能夠極大地提高效率。植物中已報(bào)道了大量的miRNAs,但蕎麥屬的尚未見報(bào)道。通過蕎麥ESTs和GSSs的嚴(yán)格篩選,共預(yù)測到13個蕎麥屬miRNAs(分屬11個miRNA家族)。預(yù)測的miRNAs在不同的植物中表現(xiàn)出保守性,miRNA家族成員表現(xiàn)出多樣性。共預(yù)測到17個潛在的靶基因,這些靶基因的功能包括代謝、生長發(fā)育、脅迫響應(yīng)、信號轉(zhuǎn)導(dǎo)等,表明miRNAs在植物生命活動中具有重要作用。miRNA家族系統(tǒng)進(jìn)化分析表明,金蕎麥的miRNAs進(jìn)化與甜蕎麥關(guān)系密切。以生物信息學(xué)方法從已知的數(shù)據(jù)庫中預(yù)測并挖掘蕎麥屬新的miRNAs及其靶基因是可行的。
[Abstract]:MicroRNA is a class of non-coding small molecules RNA. with post-transcriptional gene regulation function. It is a tedious task to discover and identify new miRNA by traditional test method. Screening miRNAs and its target genes from the database by bioinformatics can greatly improve the efficiency. A large number of miRNAs, have been reported in plants, but the genus buckwheat has not yet been reported. A total of 13 buckwheat miRNAs (belonging to 11 miRNA families) were predicted by strict screening of buckwheat ESTs and GSSs. The predicted miRNAs is conservative in different plants and the members of miRNA family are diverse. A total of 17 potential target genes were predicted. The functions of these target genes include metabolism, growth and development, stress response, signal transduction and so on, indicating that miRNAs plays an important role in plant life activities. The miRNAs evolution of buckwheat is closely related to sweet buckwheat. It is feasible to predict and mine new miRNAs and its target genes of buckwheat from known databases by bioinformatics.
