馬爾尼菲藍狀菌酵母相黑素生成和酪氨酸酶基因的關系

【摘要】:目的探討馬爾尼菲藍狀菌酵母相黑素生成和酪氨酸酶(TYR)基因的關系。方法將馬爾尼菲藍狀菌標準株分別在含和不含L-DOPA的不同培養(yǎng)基中37℃避光培養(yǎng)3天,觀察菌落形態(tài)及顏色變化,應用實時熒光定量PCR測定TYR基因mRNA的表達。結(jié)果(1)標準菌株接種在含和不含L-DOPA的SDA上菌落顏色無明顯改變,未見明顯黑素產(chǎn)生;(2)標準菌株接種在含有L-DOPA的BHI上菌落顏色深于接種在不含有L-DOPA的BHI的菌落顏色;(3)鏡下觀察標準菌株接種在含和不含L-DOPA的SDA,BHI上,其酵母樣細胞的大小、形態(tài)基本一致,均未見被染成黑色。(4)TYR基因表達:接種在含L-DOPA的BHI的標準菌株,其TYR基因表達量高于不含L-DOPA的BHI組,差異有統(tǒng)計學意義(P0.05);含和不含L-DOPA的SDA中標準菌株TYR基因表達量均低于同組BHI組,差異有統(tǒng)計學意義(P0.05)。結(jié)論馬爾尼菲藍狀菌在BHI中可以利用L-DOPA合成黑素;在BHI中添加外源性LDOPA時,馬爾尼菲藍狀菌的TYR基因表達上調(diào)。
