意大利蜜蜂6日齡幼蟲腸道內(nèi)球囊菌的差異表達基因分析

【摘要】:為檢測蜜蜂球囊菌(Ascosphaera apis)在脅迫意大利蜜蜂(Apis mellifera ligustica,簡稱意蜂)幼蟲后期的基因表達譜,利用RNA-seq技術(shù)對純培養(yǎng)的球囊菌孢子及球囊菌脅迫的意蜂6日齡幼蟲腸道進行深度測序。通過比較處理組和對照組得到21 361個差異表達基因(DEGs),包括15 306個上調(diào)基因和6 055個下調(diào)基因。GO富集分析結(jié)果顯示,上調(diào)基因富集于39個GO條目(term),基因富集數(shù)最多的為細胞進程(2 416unigenes)、細胞(2 408 unigenes)和細胞組件(2 408 unigenes);下調(diào)基因富集于38個GO term,基因富集數(shù)最多的為代謝進程(1 227 unigenes)、細胞進程(1 185 unigenes)和細胞(1 131 unigenes)。KEGG代謝通路富集分析結(jié)果顯示,上調(diào)基因富集在119個通路,其中基因富集數(shù)最多的是核糖體(221 unigenes),其次為內(nèi)質(zhì)網(wǎng)中蛋白加工(190 unigenes)和內(nèi)吞作用(177 unigenes);下調(diào)基因富集在113個通路上,基因富集數(shù)最多的是核糖體(144 unigenes),其次為RNA轉(zhuǎn)運(113 unigenes)和氨基酸生物合成(108 unigenes)。進一步分析發(fā)現(xiàn),富集在MAPK信號通路上的64個基因上調(diào)表達,說明該通路在球囊菌脅迫后期被激活。研究結(jié)果不僅為揭示球囊菌在脅迫意蜂幼蟲后期的病原-宿主互作提供了重要信息,也為闡明球囊菌致病的分子機制奠定了基礎(chǔ)。
[Abstract]:In order to detect the gene expression profile of (Ascosphaera apis) in the late larval stage of Italian honeybee (Apis mellifera ligustica, (Apis mellifera), The in-depth sequencing of the 6-day-old larvae of S.aeruginosa under pure culture and 6-day-old stress was carried out by RNA-seq technique. 21,361 differentially expressed genes (DEGs), including 15,306 up-regulated genes and 6,055 down-regulated genes, were obtained from comparison treatment group and control group. Go enrichment analysis showed that the up-regulated genes were enriched in 39 GO entry (term),. The most abundant genes were cell process (2 416unigenes), cell (2 408 unigenes) and cell module (2 408 unigenes);). The down-regulated genes were the most abundant in 38 GO term, genes (1 227 unigenes), cell process (1 185 unigenes) and 1 131 unigenes). The results of KEGG metabolic pathway enrichment analysis showed that the up-regulated genes were enriched in 1 227 unigenes cells and 1 131 unigenes cells, and the up-regulated genes were enriched in 1 227 unigenes cells and 1 131 unigenes cells. The most abundant genes were ribosomes (221 unigenes),) followed by endoplasmic reticulum protein processing (190 unigenes) and endocytosis (177 unigenes);). The down-regulated genes were enriched in 113 pathways, and the most abundant genes were ribosomes (144 unigenes), followed by RNA transport (113 unigenes) and amino acid biosynthesis (108 unigenes). Further analysis showed that 64 genes enriched in the MAPK signaling pathway were up-regulated, indicating that the pathway was activated in the late stage of stress. The results not only provide important information for revealing the pathogen-host interaction in the late stage of stress on Italian bee larvae, but also lay a foundation for elucidating the molecular mechanism of the pathogenesis of B.aeruginosa.
