【摘要】：Dof(DNA-binding one zinc finger)基因家族是植物特有的一類包含高度保守的C_2-C_2鋅指結(jié)構(gòu)域的轉(zhuǎn)錄因子,其在植物種子發(fā)育與萌發(fā)、光、激素以及逆境響應(yīng)中起到重要調(diào)控作用。本研究基于已公布的粗山羊草基因組數(shù)據(jù),采用生物信息學(xué)方法鑒定了粗山羊草Dof基因,并對其結(jié)構(gòu)、保守結(jié)構(gòu)域、染色體定位、系統(tǒng)進(jìn)化樹和表達(dá)譜進(jìn)行了詳細(xì)的分析。結(jié)果表明:粗山羊草Dof轉(zhuǎn)錄因子家族包含10個基因,它們均含有完整的C_2-C_2鋅指保守結(jié)構(gòu)域;每個基因含有1~3個內(nèi)含子,AetDof家族蛋白共包含11個保守的模體;該家族基因除7號染色體外,在其余染色體上均有分布;與擬南芥Dof蛋白構(gòu)建系統(tǒng)發(fā)育樹發(fā)現(xiàn),該家族基因可被分為3個亞族;轉(zhuǎn)錄組數(shù)據(jù)分析顯示,粗山羊草Dof基因的表達(dá)具有組織特異性,其中3個轉(zhuǎn)錄因子(AetDof1,AetDof7,AetDof10)在種子中特異性強(qiáng)表達(dá)。本研究結(jié)果為深入解析粗山羊草Dof基因的功能提供了參考依據(jù)。
[Abstract]:The DOF (DNA-binding one zinc finger) gene family is a kind of transcription factor which contains highly conserved C _ 2-C _ 2 zinc finger domain, which plays an important role in the development and germination of plant seeds, light, hormone and stress response. Based on the published genome data of Leymus chinensis, the Dof gene of Leymus chinensis is identified by means of bioinformatics, and its structure, conserved domain, chromosome location, phylogenetic tree and expression profile are analyzed in detail. The results showed that the DOF transcription factor family contained 10 genes, all of which contained a complete C _ 2-C _ 2 zinc finger conserved domain; each gene contained 1 to 3 introns, and the AetDof family protein contained 11 conservative phantom; the family gene, except for chromosome 7, It is found that the gene of the family can be divided into 3 subfamilies and 3 transcription factors (AetDof1, AetDof7, AetDof10) was strongly expressed in the seeds. The results of this study provide a reference for the in-depth analysis of the function of the Dof gene in the Leymus chinensis.
