IMPDH基因克

【摘要】:旨在獲得牦牛IMPDH基因序列并進(jìn)行生物信息學(xué)分析,同時(shí)分析其在卵巢組織中的時(shí)序表達(dá)譜,為進(jìn)一步研究卵巢生長(zhǎng)發(fā)育及排卵機(jī)制奠定基礎(chǔ)。通過(guò)RT-PCR技術(shù)克隆牦牛的IMPDH基因,并運(yùn)用生物信息學(xué)方法分析不同物種序列進(jìn)化關(guān)系,利用熒光定量qRT-PCR技術(shù)檢測(cè)IMPDH基因在不同發(fā)育時(shí)期牦牛卵巢組織中的表達(dá)情況。結(jié)果顯示,克隆得到牦牛1 623bp的IMPDH基因cDNA序列,內(nèi)含1 545bp開放閱讀框序列,編碼514個(gè)氨基酸殘基。與野牦牛的相應(yīng)序列具有很高的同源性(99.4%),表明IMPDH基因在進(jìn)化過(guò)程中相對(duì)保守。實(shí)時(shí)定量RT-PCR檢測(cè)結(jié)果顯示IMPDH基因的表達(dá)隨年齡的增加而呈現(xiàn)上升趨勢(shì)。熒光定量qRT-PCR檢測(cè)結(jié)果顯示IMPDH基因在卵巢卵泡期表達(dá)最強(qiáng),顯著高于紅體形成期。結(jié)果表明IMPDH參與牦牛卵巢組織的生長(zhǎng)發(fā)育,并在卵巢活動(dòng)中有明顯的表達(dá)規(guī)律,表明IMPDH基因在牦牛繁殖周期具有重要作用,為進(jìn)一步研究牦牛IMPDH基因功能奠定基礎(chǔ)。
[Abstract]:The aim of this study was to obtain the sequence of yak IMPDH gene and to analyze the sequence of its expression in ovarian tissue by bioinformatics analysis, so as to lay a foundation for further study of ovarian growth and ovulation mechanism. The IMPDH gene of yak was cloned by RT-PCR, and the evolutionary relationship of different species was analyzed by bioinformatics. The expression of IMPDH gene in yak ovary was detected by fluorescence quantitative qRT-PCR technique. The results showed that the cDNA sequence of IMPDH gene of yak 1 623bp was cloned, which contained 1 545bp open reading frame sequence and encoded 514 amino acid residues. There was a high homology (99.4%) with the corresponding sequence of wild yak, indicating that the IMPDH gene was relatively conserved in the course of evolution. Real-time quantitative RT-PCR analysis showed that the expression of IMPDH gene increased with age. The results of fluorescence quantitative qRT-PCR showed that the expression of IMPDH gene was the strongest in follicular stage, and was significantly higher than that in erythropoiesis stage. The results showed that IMPDH was involved in the growth and development of yak ovary and had obvious expression rule in ovarian activity. The results showed that IMPDH gene played an important role in the reproductive cycle of yak, which laid a foundation for further study on the function of yak IMPDH gene.
