【摘要】：【目的】克隆茶樹(shù)甲羥戊酸激酶基因(Cs MVK),分析其生物學(xué)信息及在不同組織及烏龍茶做青過(guò)程中的表達(dá)特性,為研究茶葉加工過(guò)程中香氣形成機(jī)制提供參考�！痉椒ā縍T-PCR擴(kuò)增Cs MVK基因,應(yīng)用生物信息學(xué)軟件對(duì)其編碼蛋白(Cs MVK)的氨基酸序列進(jìn)行預(yù)測(cè)分析,通過(guò)實(shí)時(shí)熒光定量PCR(q PCR)分析Cs MVK在茶樹(shù)不同組織及烏龍茶做青過(guò)程中的表達(dá)特性�！窘Y(jié)果】克隆獲得的Cs MVK基因(Gen Bank登錄號(hào)MF668187)全長(zhǎng)1455 bp,開(kāi)放閱讀框(ORF)長(zhǎng)度為1164 bp,編碼387個(gè)氨基酸,蛋白質(zhì)分子量41.18 k D,理論等電點(diǎn)(p I)5.88。Cs MVK蛋白具有GHMP_kinases_N domain和GHMP_kinases_C domain 2個(gè)功能結(jié)構(gòu)域,定位于細(xì)胞質(zhì)中,不存在跨膜結(jié)構(gòu)和信號(hào)肽。系統(tǒng)發(fā)育進(jìn)化樹(shù)分析結(jié)果顯示,Cs MVK與三七、常春藤的MVK聚為一類,表明茶樹(shù)與三七、常春藤的親緣關(guān)系最近。q PCR檢測(cè)結(jié)果顯示,Cs MVK基因在果實(shí)中的表達(dá)量顯著高于其他組織(P0.05,下同),在不同組織中的表達(dá)量排序?yàn)?果實(shí)根葉莖花;在烏龍茶做青過(guò)程中,從鮮葉到曬青葉,Cs MVK基因的表達(dá)量上升,曬青到一搖過(guò)程中表達(dá)量下降,經(jīng)過(guò)3次搖青,其表達(dá)量持續(xù)升高并在殺青前達(dá)最大值,與其他做青階段差異顯著。【結(jié)論】Cs MVK基因表達(dá)與茶葉香氣品質(zhì)形成有關(guān)。
[Abstract]:[objective] to clone the methylvalerate kinase gene (Cs MVK),) from tea plants and analyze its biological information and expression characteristics in different tissues and Oolong tea. [methods] the Cs MVK gene was amplified by RT-PCR and the amino acid sequence of its encoded protein (Cs MVK) was predicted by bioinformatics software. The expression characteristics of Cs MVK in different tissues of tea plant and oolong tea were analyzed by real-time fluorescence quantitative PCR (q PCR). [results] the total length of Cs MVK gene (Gen Bank accession number (MF668187) was 1455 bp,. Open reading frame (ORF) encodes 387 amino acids with a molecular weight of 41.18kD and a length of 1164 bp,. The theoretical isoelectric point (p I) 5.88.Cs MVK protein has two functional domains, GHMP_kinases_N domain and GHMP_kinases_C domain. Located in the cytoplasm, there was no transmembrane structure or signal peptide. Phylogenetic tree analysis showed that the MVK of, Cs MVK and Panax notoginseng and Ivy were clustered together, indicating that the phylogenetic relationship between tea and Panax notoginseng and ivy was recently detected by. Q PCR. The expression of Cs MVK gene in fruit was significantly higher than that in other tissues (P0.05, the same below), and the order of expression in different tissues was as follows: fruit, root, leaf, stem and flower; In the process of oolong tea green, the expression of, Cs MVK gene increased from fresh leaf to sun-green leaf, and decreased during sun-green to first shake. After three times of shaking green, the expression of, Cs MVK gene increased continuously and reached the maximum before killing green. [conclusion] the expression of Cs MVK gene is related to the formation of tea aroma quality.
