亞洲玉米螟滯育期間耐寒相關(guān)基因的表達(dá)分析

【摘要】:亞洲玉米螟Ostrrinia furnacalis(Guenee),是我國主要玉米害蟲,屬典型的兼性滯育昆蟲,通常以5齡低溫耐受型幼蟲滯育越冬。為探索亞洲玉米螟滯育幼蟲受到低溫脅迫時(shí),耐寒相關(guān)基因的表達(dá)量變化規(guī)律,本文在評估篩選亞洲玉米螟越冬幼蟲及不同蟲態(tài)內(nèi)參基因表達(dá)穩(wěn)定性的基礎(chǔ)上,選取編碼耐寒相關(guān)蛋白的基因—精氨酸激酶基因(AK),谷胱甘肽轉(zhuǎn)移酶基因(Gst)和磷酸甘油醛異構(gòu)酶基因(Tpi),研究熱馴化(20℃)與冷馴化(5℃)維持滯育狀態(tài)下及在越冬期間的滯育幼蟲的AK、Gst和Tpi基因表達(dá)的時(shí)間動(dòng)態(tài),旨在為解析亞洲玉米螟幼蟲對低溫脅迫的分子適應(yīng)機(jī)制奠定基礎(chǔ)。全文試驗(yàn)結(jié)果如下:(1)篩選出可用于研究亞洲玉米螟滯育時(shí)基因表達(dá)研究的最適內(nèi)參基因。亞洲玉米螟不同蟲態(tài)下,4個(gè)候選內(nèi)參基因的穩(wěn)定性大小排序?yàn)?8S rRNAEF1-αβ-ctinPS3;越冬滯育幼蟲中,穩(wěn)定性大小排序?yàn)棣?aactinEF1-α18S rRNARPS3。在亞洲玉米螟不同蟲態(tài)和越冬滯育幼蟲中,18SrRNA和β-actin可分別作為目的基因表達(dá)水平分析試驗(yàn)中的內(nèi)參基因。(2)探索亞洲玉米螟滯育相關(guān)的脅迫耐受性過程。分析AK、Gst和Tpi基因在確定滯育、熱馴化(20℃)與冷馴化(5℃)滯育維持過程中的表達(dá)動(dòng)態(tài)。與非滯育組相比,滯育起始階段Gst基因相對表達(dá)量顯著下調(diào),AK和Tpi基因表達(dá)量無顯著差異。在熱馴化期間,僅Tpi基因表達(dá)量下調(diào);冷馴化期間,3個(gè)基因表達(dá)量均上調(diào),且呈逐漸上升的趨勢。低溫滯育期間mRNA的半衰期延長,mRNA轉(zhuǎn)錄逐漸積累,滯育過程和溫度均影響亞洲玉米螟脅迫相關(guān)基因的表達(dá)。(3)了解越冬期間滯育幼蟲的AK、Gst和Tpi基因表達(dá)的時(shí)間動(dòng)態(tài)。分析越冬滯育幼蟲AK、Gst和Tpi基因的表達(dá)量,亞洲玉米螟幼蟲越冬期間AK、Gst和Tpi基因表達(dá)量均呈上調(diào),且與越冬環(huán)境溫度呈負(fù)相關(guān)性。表明能量代謝和抗氧化反應(yīng)作用于亞洲玉米螟滯育幼蟲的低溫脅迫過程。
