黃連素調(diào)節(jié)BMP4轉(zhuǎn)錄通路基因表達(dá)改善2型糖尿病地鼠內(nèi)臟白色脂肪組織胰島素抵抗的研究

【摘要】:研究黃連素對2型糖尿病地鼠內(nèi)臟白色脂肪組織(VWAT)中BMP4轉(zhuǎn)錄通路和白/棕脂組織轉(zhuǎn)換通路基因mRNA表達(dá)的影響并探討相關(guān)機(jī)制。應(yīng)用高脂飲食誘導(dǎo)肥胖胰島素抵抗地鼠模型,給予小劑量鏈脲菌素建立2型糖尿病地鼠模型。造模結(jié)束后隨機(jī)分成正常對照組、肥胖胰島素抵抗組、2型糖尿病組和2型糖尿病黃連素治療組。治療9周后,應(yīng)用實時定量PCR方法檢測各組地鼠VWAT中BMP4通路、白/棕脂組織轉(zhuǎn)換通路及靶基因的mRNA表達(dá)改變。結(jié)果顯示,模型地鼠VWAT中BMP4,BMPRⅡ,BMPRl A,Smad1,Smad5,Smad8,p38/MAPK,ATF2,PRDM16,C/EBPβ,PGC1α,PPARγ及棕脂組織特異基因的mRNA表達(dá)降低,而Smad6,Smurf1和白脂組織特異基因的mRNA表達(dá)增加。黃連素治療調(diào)節(jié)VWAT中BMP4轉(zhuǎn)錄通路和棕脂組織轉(zhuǎn)錄通路,誘導(dǎo)棕脂組織特異基因mRNA的表達(dá),誘導(dǎo)白色脂肪棕色化基因表型,改善脂誘性胰島素抵抗。結(jié)果表明,BMP4轉(zhuǎn)錄通路參與脂誘性內(nèi)臟白色脂肪組織胰島素抵抗(FIVWATIR)的形成及黃連素誘導(dǎo)白色脂肪棕色化的分子機(jī)制。
[Abstract]:To study the effect of berberine on the expression of BMP4 transcriptional pathway and gene mRNA in white adipose tissue of visceral white adipose tissue of type 2 diabetic hamster, and to explore the related mechanism. The obese insulin resistant hamster model was induced by high fat diet and the type 2 diabetic hamster model was established by low dose streptozotocin. The model was randomly divided into normal control group, obese insulin resistance group, type 2 diabetes group and type 2 diabetic berberine treatment group.
