油葵HaFAD2-2基因的克隆及表達(dá)分析

【摘要】:脂肪酸脫氫酶2(fatty acid desaturase,FAD2)催化油酸生成亞油酸,是植物體內(nèi)生成多不飽和脂肪酸的關(guān)鍵酶。根據(jù)已報(bào)道的向日葵(Helianthus annuus L.)FAD2基因序列,設(shè)計(jì)引物進(jìn)行RT-PCR,克隆得到油葵FAD2-2基因全長(zhǎng)cDNA,命名為HaFAD2-2。該基因開(kāi)放閱讀框?yàn)? 152bp,編碼383個(gè)氨基酸,相對(duì)分子質(zhì)量43.96kD,等電點(diǎn)為8.56。對(duì)基因組進(jìn)行內(nèi)含子調(diào)查發(fā)現(xiàn),該基因在編碼區(qū)內(nèi)沒(méi)有內(nèi)含子。多序列比對(duì)和系統(tǒng)進(jìn)化分析發(fā)現(xiàn),FAD2-2基因編碼蛋白與金盞菊(Calendula officinalis)、斑鳩菊(Vernonia galamensis)等菊科植物具有較近的親緣關(guān)系。qRT-PCR分析表明,HaFAD2-2基因在根、莖、葉、花、子葉和未成熟種子中均有表達(dá),且以葉中的表達(dá)量最高,未成熟種子中的表達(dá)量最低;低溫(5℃、15℃)脅迫處理能顯著促進(jìn)該基因在根中的表達(dá),抑制其在葉中的表達(dá);鹽脅迫(300 mmol/L NaCl)處理對(duì)其表達(dá)也具有抑制作用。該研究結(jié)果可為進(jìn)一步探討HaFAD2-2基因的功能奠定基礎(chǔ)。
[Abstract]:Fatty acid dehydrogenase (2 (fatty acid desaturase,FAD2) catalyzes oleic acid to produce linoleic acid, which is the key enzyme to produce polyunsaturated fatty acids in plants. According to the reported sequence of sunflower (Helianthus annuus L.) FAD2 gene, a primer was designed for RT-PCR, cloning to obtain the full length cDNA, of FAD2-2 gene of oil sunflower named HaFAD2-2.. The open reading frame of the gene is 1152 BP, encoding 383 amino acids, the relative molecular weight is 43.96 kD and the isoelectric point is 8.56. An intron investigation of the genome revealed that the gene had no intron in the coding region. Multiple sequence alignment and phylogenetic analysis showed that the FAD2-2 gene encoded protein was closely related to (Calendula officinalis), (Vernonia galamensis) and other plants of Compositae. QRT-PCR analysis showed that HaFAD2-2 gene was found in roots, stems, leaves and flowers. Both cotyledons and immature seeds had the highest expression, and the lowest in immature seeds.
