【摘要】：以溫敏型黃瓜‘C09-123’為試材,通過對不同夜溫處理的黃瓜幼葉進行差異蛋白質(zhì)鑒定,篩選出一個與低夜溫影響黃瓜雌性形成密切相關(guān)的蛋白,采用生物信息學(xué)方法,對該蛋白進行生物信息學(xué)分析,并運用基因克隆技術(shù)克隆了該基因,以期為明確該基因在黃瓜性別分化中的作用機制奠定基礎(chǔ),也為進一步試驗研究提供基因材料。結(jié)果表明:該基因全長870bp,編碼289個氨基酸,該基因編碼的蛋白質(zhì)屬于葉綠體中一個不穩(wěn)定的疏水蛋白質(zhì),無明顯信號肽,無跨膜結(jié)構(gòu)。與甜瓜為同一進化分支,同源性較高。該蛋白屬于PAPfibrillin蛋白家族,因此將編碼該蛋白的基因命名為CsPAP-fib。
[Abstract]:Using 'C09-123', a thermo-sensitive cucumber as material, a protein closely related to the effect of low night temperature on female formation of cucumber was screened by differential protein identification of young cucumber leaves treated with different night temperature. The bioinformatics method was used to study the effect of low night temperature on the female formation of cucumber. The protein was analyzed by bioinformatics and cloned by gene cloning technique in order to lay a foundation for clarifying the function mechanism of the gene in cucumber sex differentiation and to provide genetic materials for further experimental study. The results showed that the gene was 870 BP in length, encoding 289 amino acids. The protein encoded by the gene belonged to an unstable hydrophobic protein in chloroplast, with no obvious signal peptide and no transmembrane structure. It is the same evolutionary branch as muskmelon and has high homology. The protein belongs to the PAPfibrillin protein family, so the gene that encodes the protein is named CsPAP-fib.
