擬南芥At5g66070基因在ABA處理下的初步研究

【摘要】:本研究以擬南芥為實驗材料,探索基因At5g66070在植物激素ABA處理條件下的相關(guān)生理功能.結(jié)果表明,在10μMABA處理下,該基因的表達(dá)量明顯升高,表明該基因受到ABA的誘導(dǎo).亞細(xì)胞定位發(fā)現(xiàn)AT5G66070定位在細(xì)胞核.通過DNA及RNA水平篩選鑒定At5g66070基因T-DNA插入純合突變體,然后經(jīng)根瘤農(nóng)桿菌介導(dǎo)法進(jìn)行遺傳轉(zhuǎn)化,篩選獲得過表達(dá)轉(zhuǎn)基因株系.表型分析發(fā)現(xiàn)經(jīng)ABA處理后,突變體相對于野生型而言根長較短,表明突變體對ABA更為敏感.氣孔關(guān)閉實驗發(fā)現(xiàn)10μM ABA能誘導(dǎo)突變體氣孔關(guān)閉,而野生型和過表達(dá)無顯著影響.以上結(jié)果表明At5g66070在擬南芥的ABA脅迫響應(yīng)中起負(fù)調(diào)控作用.
[Abstract]:In this study, Arabidopsis thaliana was used as experimental material to explore the physiological function of gene At5g66070 under the condition of plant hormone ABA treatment. The results showed that the expression of the gene was significantly increased under the treatment of 10 渭 MABA, indicating that the gene was induced by ABA. Subcellular localization revealed that AT5G66070 was located in the nucleus. The At5g66070 gene T-DNA was inserted into homozygous mutants by DNA and RNA screening, and then transformed by Agrobacterium tumefaciens. Phenotypic analysis showed that the root length of the mutant was shorter than that of the wild type after ABA treatment, indicating that the mutant was more sensitive to ABA. Stomatal closure test showed that 10 渭 M ABA could induce stomatal closure of mutant, but wild type and overexpression had no significant effect. These results suggest that At5g66070 plays a negative role in the response to ABA stress in Arabidopsis thaliana.
