【摘要】：目的研究PJA1及miR-130a在口腔腫瘤組織中的表達情況,明確對腫瘤細胞遷移增殖的影響,并探究兩種基因之間的相互作用。方法收集15組口腔腫瘤及其瘤旁組織,提取組織內(nèi)RNA,應(yīng)用qRT-PCR法檢測組織中PJA1及miR-130a的表達情況;構(gòu)建PJA1過表達、miR-130a過表達及兩者同時過表達的口腔腫瘤細胞株SCC-3,應(yīng)用MTT實驗和Wound scratch assay實驗分別檢測細胞增殖與遷移的影響。結(jié)果 qRT-PCR結(jié)果表明PJA1在口腔腫瘤組織中的表達明顯低于瘤旁組織(P0.05),miR-130a在口腔腫瘤組織中的表達明顯高于瘤旁組織(P0.05);MTT和Wound scratch assay實驗結(jié)果表明PJA1過表達可以抑制腫瘤細胞的增殖與遷移,miR-130a過表達可以促進腫瘤細胞的增殖與遷移,兩者同時過表達PJA1可以抑制miR-130a促進腫瘤細胞增殖與遷移的能力。結(jié)論 PJA1在口腔腫瘤組織中低表達,具有抑制腫瘤細胞增殖和遷移的能力;miR-130a在口腔腫瘤組織中高表達,具有促進腫瘤細胞增殖和遷移的能力;miR-130a可能通過抑制了PJA1從而促進腫瘤細胞的增殖和遷移。
[Abstract]:Objective to study the expression of PJA1 and miR-130a in oral tumor tissues, to investigate the effects of PJA1 and miR-130a on the migration and proliferation of tumor cells and to explore the interaction between the two genes. Methods the expressions of PJA1 and miR-130a in 15 groups of oral tumors and their adjacent tissues were detected by qRT-PCR method and RNA, was extracted from the tissues. Overexpression of PJA1, overexpression of miR-130a and overexpression of both in oral tumor cell line SCC-3, were used to detect the effect of cell proliferation and migration by MTT assay and Wound scratch assay, respectively. Results qRT-PCR results showed that the expression of PJA1 in oral tumor tissue was significantly lower than that in adjacent tumor tissue (P0.05), and the expression of miR-130a in oral tumor tissue was significantly higher than that in paratumor tissue (P0.05). The results of MTT and Wound scratch assay experiments showed that the overexpression of PJA1 could inhibit the proliferation and migration of tumor cells, and the overexpression of miR-130a could promote the proliferation and migration of tumor cells. Both overexpression of PJA1 could inhibit the ability of miR-130a to promote the proliferation and migration of tumor cells. Conclusion PJA1 can inhibit the proliferation and migration of tumor cells in oral tumor tissues, and the high expression of miR-130a in oral tumor tissues can promote the proliferation and migration of tumor cells. MiR-130a may promote the proliferation and migration of tumor cells by inhibiting PJA1.
