北京及周邊地區(qū)九個(gè)地理種群的亞洲小車(chē)蝗mtDNA ND6基因及其部分側(cè)翼序列分析

【摘要】:為明確北京地區(qū)亞洲小車(chē)蝗Oedaleus asiaticus Bey-Bienko的遷飛來(lái)源,選取包含完整線(xiàn)粒體DNA的ND6、tRNA-Thr基因及側(cè)翼ND4L、CYTB基因部分序列作為目的片段,采用DNA序列法分析來(lái)自北京及周邊省市9個(gè)地理種群155個(gè)亞洲小車(chē)蝗樣本的序列堿基組成、種群遺傳多樣性、種群結(jié)構(gòu)及單倍型網(wǎng)絡(luò)進(jìn)化等。結(jié)果表明:在目的序列中共檢測(cè)到多態(tài)性位點(diǎn)16個(gè),鑒定出單倍型20種,分別占?jí)A基總數(shù)和總樣品量的1.88%和12.90%;核苷酸多樣性為0.0012,單倍型多樣性為0.66,種群間遺傳分化系數(shù)絕對(duì)值≤0.51,種群內(nèi)個(gè)體間遺傳變異占總變異的86.88%,各種群遺傳變異與空間距離間無(wú)顯著相關(guān)性,供試樣本總體上遺傳變異程度相對(duì)較低。北京密云種群鑒定出的獨(dú)享單倍型H14由單倍型H10經(jīng)2次突變獲得,H10被錫林郭勒和烏蘭察布種群共享;北京密云與河北灤平、圍場(chǎng)及內(nèi)蒙古烏蘭察布種群間基因流Nm絕對(duì)值分別為3.749、1.387和0.912,均高于密云種群與其它種群間的Nm絕對(duì)值;結(jié)合亞洲小車(chē)蝗在北京及周邊地區(qū)出現(xiàn)的先后時(shí)序可知,內(nèi)蒙古錫林郭勒、烏蘭察布及河北灤平、圍場(chǎng)是北京地區(qū)亞洲小車(chē)蝗的最主要遷飛來(lái)源。
[Abstract]:In order to identify the migration source of Oedaleus asiaticus Bey-Bienko, the partial sequences of ND6,tRNA-Thr gene and flanking ND4L,CYTB gene containing intact mitochondrial DNA were selected as the target fragments. DNA sequence method was used to analyze the sequence base composition, population genetic diversity, population structure and haplotype network evolution of 155 Asian locust samples from 9 geographical populations in and around Beijing. The results showed that there were 16 polymorphic loci detected in the target sequence, and 20 haplotypes were identified, accounting for 1.88% of the total number of bases and 12.90% of the total samples.
