胸腺移植合并CD4~--DLI對(duì)異基因造血干細(xì)胞移植后T細(xì)胞重建的影響

【摘要】:目的研究胸腺移植(TT)合并CD4~--DLI對(duì)異基因造血干細(xì)胞移植(allo-HSCT)后T細(xì)胞重建的影響。方法將BALB/c小鼠隨機(jī)分成3組,單純?cè)煅杉?xì)胞移植(HSCT組),造血干細(xì)胞移植+胸腺移植(TT組),造血干細(xì)胞移植~+胸腺移植+去CD4~+T細(xì)胞的供體淋巴細(xì)胞輸注(CD4~--DLI組)。移植前1 d,對(duì)小鼠進(jìn)行致死劑量的放療處理。移植當(dāng)天,進(jìn)行造血干細(xì)胞移植,供體為C57BL/6小鼠。移植處理后5 d、2周、4周、3個(gè)月分別處死小鼠,取小鼠外周血和脾臟細(xì)胞進(jìn)行流式細(xì)胞術(shù)檢測(cè)T細(xì)胞表面抗原,T細(xì)胞受體,初始T細(xì)胞,細(xì)胞內(nèi)因子;HE染色觀察供體胸腺的發(fā)育情況。結(jié)果 TT和CD4~--DLI不會(huì)影響彼此對(duì)T細(xì)胞重建的作用;TT合并CD4~--DLI能夠通過胸腺依賴和胸腺非依賴兩種途徑加快CD4~+和CD8~+T細(xì)胞數(shù)目的重建、擴(kuò)大初始CD4~+和CD8~+T細(xì)胞庫、加快T細(xì)胞功能的重建,并且對(duì)TCR庫和胸腺?zèng)]有副作用。結(jié)論:TT合并CD4~--DLI可以加快allo-HSCT后T細(xì)胞重建。
[Abstract]:Objective to study the effect of thymus transplantation (TT) combined with CD4~--DLI on T cell reconstruction after allogeneic hematopoietic stem cell transplantation (allo-HSCT). Methods BALB/c mice were randomly divided into three groups: simple hematopoietic stem cell transplantation (HSCT group), hematopoietic stem cell transplantation thymus transplantation (TT group). Hematopoietic stem cell transplantation ~ thymus transplantation donor lymphocyte infusion of CD4~ T cells (CD4~--DLI group). The mice were treated with lethal dose of radiotherapy one day before transplantation. On the day of transplantation, hematopoietic stem cells were transplanted to C57BL/6 mice. Mice were killed 5 days after transplantation for 2 weeks, 4 weeks and 3 months respectively. Peripheral blood and spleen cells were collected for flow cytometry to detect T cell surface antigen, T cell receptor, initial T cell and intracellular factor. The development of donor thymus was observed by HE staining. Results TT and CD4~--DLI had no effect on T cell reconstruction. TT combined with CD4~--DLI can accelerate the reconstruction of CD4~ and CD8~ T cells by thymus dependence and thymus independence, expand the initial CD4~ and CD8~ T cell banks, and accelerate T cell function reconstruction, and has no side effects on TCR library and thymus. Conclusion: TT combined with CD4~--DLI can accelerate T cell reconstruction after allo-HSCT.
