4個線粒體基因在鯉所屬國家種質(zhì)鑒定中的應(yīng)用及效果評價

【摘要】:近年來,mtDNA條形碼技術(shù)已廣泛應(yīng)用于魚類種質(zhì)鑒定研究。為探討該技術(shù)在鯉種質(zhì)鑒定方面應(yīng)用的可行性及如何有效應(yīng)用于地域種群等鑒定分析,從NCBI網(wǎng)站(http://www.ncbi.nlm.nih.gov/)搜集鯉COⅠ、COⅡ、Cytb和D-loop4個mtDNA序列,并嘗試基于DNA條形碼技術(shù)來進(jìn)行鯉所屬國家的鑒定分析。結(jié)果表明:Cytb相對其他3個線粒體序列存在較大的DNA條形碼間隙,COⅡ基因最佳匹配的準(zhǔn)確率最高(準(zhǔn)確率82.97%),基于4個mtDNA序列的聚類分析表明鯉所屬國家按照臨近區(qū)域聚類。綜合條形碼間隙及假陽性率和假陰性率的評價,研究認(rèn)為,相比其他3個mtDNA序列,Cytb基因用于鑒定鯉所屬國家更具優(yōu)勢,但其鑒定效率仍不理想。綜上所述,mtDNA序列用于鑒定鯉不同地域的技術(shù)依然需要改進(jìn),以降低這一技術(shù)的假陽性率和假陰性率。
