刀鱭POMC基因的cDNA克隆及其應(yīng)激應(yīng)答

【摘要】:為了研究刀鱭(Coilia nasus)阿黑皮素原(proopiomelanocortin,POMC)基因的應(yīng)激應(yīng)答及調(diào)控,克隆了刀鱭POMC基因的c DNA全長。刀鱭POMC基因的c DNA全長2030 bp,編碼區(qū)699 bp,編碼232個氨基酸。二級結(jié)構(gòu)預測顯示,刀鱭POMC包括信號肽(signal peptide)、N末端區(qū)(N-terminal region,NPP)、促腎上腺皮質(zhì)激素結(jié)構(gòu)域(adrenocorticotropic hormone,ACTH)、β-脂解素(β-LPH)、α-促黑素(α-MSH)、促腎上腺皮質(zhì)激素樣垂體中葉多肽(CLIP)、γ-脂解素(γ-LPH)、β-促黑素(β-MSH)和β-內(nèi)咖肽(β-EP)結(jié)構(gòu)域。運用熒光定量的方法檢測了該基因在刀鱭不同組織中的表達分布。結(jié)果顯示,POMC在健康刀鱭的腦高表達,鰓、腎、精巢相對高表達,肝、脾、腸、頭腎、肌肉和卵巢中微量表達。運輸脅迫后,POMC基因的表達量顯著下降(P0.05);10‰ NaCl組4 h和6 h POMC基因的表達量顯著上調(diào)(P0.05),6 h的表達量恢復至應(yīng)激前。POMC基因在脂肪代謝及應(yīng)激調(diào)控中發(fā)揮重要的功能,本研究結(jié)果可為該基因在刀鱭后續(xù)育種及應(yīng)激調(diào)控工作提供重要基礎(chǔ)。
[Abstract]:In order to study the stress response and regulation of the proopiomelanocortin,POMC gene of Coilia ectenes, the full length of c DNA of the POMC gene of Coilia ectenes was cloned. The total length of c DNA of Coilia ectenes POMC gene is 2030 bp, and 699 bp, encodes 232 amino acids. The secondary structure prediction shows that Coilia ectenia POMC includes signal peptide (signal peptide), N terminal region (N-terminal region,NPP), adrenocorticotropic hormone domain (adrenocorticotropic hormone,ACTH) and 尾-LPH), 偽-melanotropin (偽-MSH),). The domain of (CLIP), 緯 -lipopolysaccharide (緯-LPH), 尾-melanin (尾-MSH) and 尾-endogenous peptide (尾-EP) in adrenocorticotropin-like pituitary gland. The expression and distribution of this gene in different tissues of Coilia ectenes were detected by fluorescence quantitative method. The results showed that the expression of POMC was high in brain, relatively high in Gill, kidney and testis, and in liver, spleen, intestine, head and kidney, muscle and ovary.
