蝴蝶蘭PhalPI基因的克隆及在花器官突變體中的表達(dá)分析

【摘要】:為深入研究蘭科植物花器官發(fā)育的調(diào)控機(jī)理,從蝴蝶蘭花瓣中克隆了一個(gè)B類MADS-box轉(zhuǎn)錄因子PhalPI(GenBank登錄號為KY020416)。序列分析表明,該基因的cDNA全長為944 bp,含完整的開放閱讀框,可編碼210個(gè)氨基酸,屬于BGLO/PI蛋白家族,與蝴蝶蘭屬的PhPI10和PeMADS6基因關(guān)系最近;表達(dá)模式分析表明,PhalPI基因在生殖器官中表達(dá),在營養(yǎng)器官中不表達(dá),在授粉后的子房中,該基因的表達(dá)水平降低。在5種花器官突變體中,PhalPI基因在萼片唇瓣化突變體的萼片和蕊柱中表達(dá)水平明顯升高;在雄蕊花瓣化突變體的萼片和側(cè)瓣中表達(dá)水平降低,在其唇瓣和蕊柱中顯著升高;在側(cè)瓣合柱化突變體的蕊柱中,PhalPI基因的表達(dá)也發(fā)生了顯著升高;PhalPI基因表達(dá)的改變與花器官形態(tài)的突變相關(guān);而在側(cè)瓣唇瓣化和側(cè)瓣花藥化突變體中,PhalPI基因的表達(dá)水平?jīng)]有變化。推測該基因在決定蝴蝶蘭側(cè)瓣和唇瓣的發(fā)育中起重要的調(diào)控作用。
[Abstract]:In order to study the regulation mechanism of flower organ development in orchids, a class B MADS-box transcription factor (PhalPI (GenBank) was cloned from the petals of Phalaenopsis (PhalPI (GenBank accession number is KY020416). Sequence analysis showed that the cDNA length of the gene was 944 bp, with a complete open reading frame, encoding 210 amino acids, belonging to the BGLO/PI protein family, and most closely related to the PhPI10 and PeMADS6 genes of Phalaenopsis. Expression pattern analysis showed that PhalPI gene was expressed in reproductive organs, but not in vegetative organs, but decreased in ovary after pollination. In the five floral mutants, the expression level of PhalPI gene was significantly increased in sepals and stamens, but decreased in the sepals and lateral petal mutants of stamen petal mutants. The expression of PhalPI gene was also significantly increased in the column of lateral valve column mutants, and the change of PhalPI gene expression was related to the mutation of floral organ morphology. However, there was no change in the expression level of PhalPI gene in the lateral valvular and lateral valve anther mutants. It is speculated that this gene plays an important role in determining the development of lateral and labial valves of Phalaenopsis.
