海南地區(qū)女性HLA-DQB1和DRB5基因多態(tài)性與宮頸癌易感性的關(guān)系

【摘要】:目的探討海南地區(qū)漢族女性人群中人類白細(xì)胞抗原(HLA)-DQB1和HLA-DRB5等位基因多態(tài)性與宮頸癌易感性的相關(guān)性。方法以海南地區(qū)41例經(jīng)病理確診的宮頸癌患者和38例因子宮肌瘤行全子宮切除術(shù)且病理證實(shí)無(wú)宮頸癌的患者為研究對(duì)象。采用PCR克隆測(cè)序技術(shù)對(duì)兩組HLA-DQB1和DRB5等位基因進(jìn)行分型。結(jié)果共檢測(cè)到12種HLA-DQB1等位基因和8種HLA-DRB5等位基因。其中HLA-DQB1*06011,HLA-DQB1*030101和HLA-DQB1*03032與HLA-DRB5*01:01:01分別是海南地區(qū)漢族女性HLA-DQB1和DRB5的優(yōu)勢(shì)表達(dá)基因。無(wú)宮頸癌患者HLA-DRB5*0206等位基因頻率(18.4%)顯著高于宮頸癌患者(0)(P=0.013)。結(jié)論 HLA-DRB5*0206可能是海南地區(qū)女性宮頸癌的保護(hù)基因,可能降低女性患宮頸癌的風(fēng)險(xiǎn)。HLA-DRB5不同等位基因可能與宮頸癌的發(fā)生存在相關(guān)性。
[Abstract]:Objective to investigate the association between polymorphisms of human leukocyte antigen (HLA) DQB1 and HLA-DRB5 alleles and susceptibility to cervical cancer in Han women in Hainan area. Methods Forty-one patients with cervical cancer confirmed by pathology and 38 patients with total hysterectomy due to uterine leiomyoma in Hainan area were studied. Two groups of HLA-DQB1 and DRB5 alleles were genotyped by PCR cloning and sequencing. Results 12 HLA-DQB1 alleles and 8 HLA-DRB5 alleles were detected. Among them, HLA-DQB1*06011,HLA-DQB1*030101, HLA-DQB1*03032 and HLA-DRB5*01:01:01 are the dominant expression genes of HLA-DQB1 and DRB5 in Han women in Hainan area, respectively. The frequency of HLA-DRB5*0206 allele in patients without cervical cancer (18.4%) was significantly higher than that in patients without cervical cancer (0) (P0. 013). Conclusion HLA-DRB5*0206 may be a protective gene for cervical cancer in Hainan women and may reduce the risk of cervical cancer in women. Different alleles of HLA-DRB5 may be associated with the occurrence of cervical cancer.
