刺參不同組織中MnSOD基因在脂多糖刺激下的定量表達(dá)分析(英文)

【摘要】:刺參由于其重要的經(jīng)濟(jì)價(jià)值和藥用價(jià)值,已經(jīng)成為我國海水養(yǎng)殖的重要種類。MnSOD對于增強(qiáng)吞噬細(xì)胞防御能力和整個(gè)機(jī)體的免疫功能具有重要作用,與生物體抵御病毒細(xì)菌等能力密切相關(guān)。通過研究在脂多糖刺激下MnSOD基因的定量表達(dá)情況,分析MnSOD在刺參免疫系統(tǒng)中的作用。結(jié)果顯示:MnSOD基因的表達(dá)量由腸、肌肉到管足逐漸降低,體壁的表達(dá)量與體腔液基本一致。當(dāng)刺參暴露于脂多糖中時(shí),除腸以外其他的四種組織中MnSOD基因均出現(xiàn)過表達(dá)。其中,體腔液和體壁組織中MnSOD基因的表達(dá)量于12 h達(dá)到峰值;肌肉和管足中MnSOD基因的表達(dá)量于48 h達(dá)到峰值;體腔液中MnSOD基因的表達(dá)量于12 h達(dá)到對照組的4 115倍,而腸組織中MnSOD基因的表達(dá)量在48 h的實(shí)驗(yàn)過程中呈波動(dòng)性降低?傮w上來說,在脂多糖刺激下刺參各組織中MnSOD基因的表達(dá)量呈規(guī)律性波動(dòng)。實(shí)驗(yàn)結(jié)果表明:在脂多糖刺激下刺參個(gè)體會產(chǎn)生強(qiáng)烈的抗氧化反應(yīng)以保護(hù)刺參免受病原微生物的感染。
[Abstract]:Because of its important economic value and medicinal value, sea cucumber has become an important species of marine culture in China. MnSOD plays an important role in enhancing phagocyte defense and immune function of the whole body. It is closely related to the ability of organism to resist virus and bacteria. The role of MnSOD in the immune system of sea cucumber was analyzed by studying the quantitative expression of MnSOD gene stimulated by lipopolysaccharide. The results showed that the expression of MnSOD gene decreased gradually from intestine, muscle to tubula, and the expression of MnSOD gene in body wall was basically consistent with that in body cavity fluid. When Acanthopsis japonicus was exposed to lipopolysaccharide, MnSOD gene was expressed in all four tissues except intestine. The expression of MnSOD gene in body cavity fluid and body wall reached the peak at 12 h, and the expression of MnSOD gene in muscle and tube foot reached the peak at 48 h. The expression of MnSOD gene in somatic cavity fluid was 4 115 times higher than that in control group at 12 h, while the expression of MnSOD gene in intestinal tissue decreased at 48 h. In general, the expression of MnSOD gene fluctuated regularly in the tissues stimulated by lipopolysaccharide. The results showed that the individuals of Acanthopsis japonicus were stimulated by lipopolysaccharide to produce strong antioxidant reactions to protect them from pathogenic microorganisms.
