【摘要】：本文通過(guò)體外細(xì)胞實(shí)驗(yàn)應(yīng)用6-姜烯酚對(duì)HCT116和HT29進(jìn)行干預(yù),以探討6-姜烯酚誘導(dǎo)結(jié)直腸癌細(xì)胞凋亡及與相關(guān)凋亡蛋白(Bax、BCL2、Caspase3和PARP1)表達(dá)之間的關(guān)系。利用倒置熒光顯微鏡觀察不同濃度的6-姜烯酚干預(yù)HCT116及HT29后細(xì)胞形態(tài)的變化,CCK8法測(cè)定不同分組HCT116及HT29抑制率,并繪制細(xì)胞增殖曲線,Annexin V-FITC/PI流式細(xì)胞術(shù)檢測(cè)不同分組HCT116及HT29凋亡,Western-blot檢測(cè)分析凋亡相關(guān)蛋白表達(dá)情況(Bax、BCL2、Caspase3和PARP1)。結(jié)果顯示,與對(duì)照組相比,6-姜烯酚可不同程度地誘導(dǎo)HCT116及HT29凋亡,促進(jìn)Bax、Caspase3和PARP1蛋白水平表達(dá)(p0.05),抑制BCL2蛋白水平表達(dá)(p0.05),并增加Bax/BCL2比值比例(p0.05)。因此,6-姜烯酚誘導(dǎo)HCT116、HT29發(fā)生凋亡可能是與激活Bax、Caspase3和PARP1的表達(dá)以及增加Bax/BCL2比值比例相關(guān),這一發(fā)現(xiàn)可為結(jié)直腸癌的臨床治療與預(yù)防提供有效參考依據(jù)。
[Abstract]:In this study, 6-gingerol was used to interfere with HCT116 and HT29 in vitro to investigate the relationship between 6-gingingenol induced apoptosis and the expression of Bax,BCL2,Caspase3 and PARP1 in colorectal cancer cells. The morphological changes of HCT116 and HT29 cells were observed by inverted fluorescence microscope. The inhibition rates of HCT116 and HT29 in different groups were measured by CCK8 method, and the cell proliferation curves were plotted. Annexin V-FITC/PI flow cytometry was used to detect HCT116 and HT29 apoptosis in different groups, and Western-blot was used to detect the expression of apoptosis-related proteins (Bax,BCL2,Caspase3 and PARP1). The results showed that 6-gingerol could induce the apoptosis of HCT116 and HT29, promote the expression of Bax,Caspase3 and PARP1 protein (p0.05), and inhibit the expression of BCL2 protein (p0.05). The ratio of Bax/BCL2 was increased (p0. 05). Therefore, the apoptosis of HCT116,HT29 induced by 6-gingerol may be related to the activation of the expression of Bax,Caspase3 and PARP1 and the increase of the ratio of Bax/BCL2, which may provide an effective reference for the clinical treatment and prevention of colorectal cancer.
