根癌農(nóng)桿菌介導(dǎo)GUS基因轉(zhuǎn)化玉米芽尖生長(zhǎng)點(diǎn)的研究

【摘要】:本試驗(yàn)以玉米(Zea mays)雜交種‘鄭單958’的芽尖生長(zhǎng)點(diǎn)作為農(nóng)桿菌侵染受體,以β-葡萄糖醛酸苷酶基因(GUS)作為轉(zhuǎn)化基因,吸水鏈霉菌草丁膦乙酰轉(zhuǎn)移酶基因(bar)作為篩選標(biāo)記基因,通過(guò)GUS顏色反應(yīng),分別從侵染液濃度、侵染時(shí)間和真空處理時(shí)間三個(gè)方面分析了影響GUS基因轉(zhuǎn)化率的因素,初步得出結(jié)論:當(dāng)侵染液濃度(OD600)為0.6、侵染時(shí)間2 h、真空處理時(shí)間為10 min時(shí),選用雜交種‘鄭單958’作為受體材料將會(huì)達(dá)到較高的遺傳轉(zhuǎn)化率。
[Abstract]:In this experiment, the growth point of shoot tip of maize (Zea mays) hybrid 'Zhengdan 958' was used as the infection receptor of Agrobacterium tumefaciens, the 尾 -glucuronidase gene (GUS) was used as the transformation gene, and the (bar) gene of Streptomyces hygroscopicus was used as the screening marker gene. By GUS color reaction, the factors affecting the conversion of GUS gene were analyzed from three aspects: the concentration of infection solution, the time of infection and the time of vacuum treatment. It is concluded that when the concentration of infection solution (OD600) is 0.6, the infection time is 2 h, and the vacuum treatment time is 10 min, the genetic transformation rate of the hybrid 'Zhengdan 958' will reach a higher genetic transformation rate when it is selected as the recipient material.
