水曲柳FmABI5基因非生物脅迫及信號誘導的表達模式分析

【摘要】:ABI5(abscisic acid-insensitive 5)蛋白是一個響應ABA信號的堿性亮氨酸拉鏈類(basic leucine zipper,b ZIP)轉錄因子。揭示了水曲柳FmABI5在非生物脅迫下的表達特征,為該基因在水曲柳中代謝調控功能的研究奠定基礎。獲得了水曲柳ABI5基因的全長,命名為FmABI5,應用生物信息學軟件分析了水曲柳FmABI5基因的分子結構特征,利用PEG、低溫(4℃)及鹽(NaCl)進行非生物脅迫處理,利用脫落酸(ABA)和赤霉素(GA3)進行信號誘導,分析FmABI5的表達特征。生物信息學分析表明該基因全長1 455 bp,含有完整的開放閱讀框,編碼484個氨基酸。FmABI5為不穩(wěn)定類親水性蛋白,不存在信號肽,具有跨膜能力,α-螺旋、延伸鏈、無規(guī)則卷曲分布于整個蛋白。分子進化分析結果表明,水曲柳FmABI5基因與芝麻的遺傳距離較近,說明其親緣關系較近;與馬鈴薯、潘那利番茄、水茄與甜椒的遺傳距離較遠,說明其親緣關系較遠。非生物脅迫結果表明,FmABI5基因表達水平隨非生物脅迫處理時間不同而上下波動,但在處理6、48和72 h后,FmABI5基因對3種非生物脅迫處理都上調表達,說明FmABI5基因對非生物脅迫具有響應。信號誘導結果表明,外源的ABA與GA共同調節(jié)了FmABI5基因的表達。
