基因芯片技術(shù)研究黃秦艽對(duì)雪上一枝蒿急性心臟毒性的作用機(jī)制

【摘要】:目的:利用芯片技術(shù)探討黃秦艽對(duì)雪上一枝蒿所致小鼠心臟急性毒性基因表達(dá)的影響及分子機(jī)制.方法:25只雄性昆明小鼠隨機(jī)分為正常對(duì)照組、雪上一枝蒿氯仿部位組(CFA)和黃秦艽水提物組(WVBF),通過(guò)表達(dá)譜對(duì)各組心臟差異表達(dá)基因進(jìn)行生物信息學(xué)研究.結(jié)果:WVBF對(duì)CFA誘導(dǎo)的急性心臟毒性具有明顯的解毒效果,染毒組與正常組相比,差異基因的功能主要涉及機(jī)體內(nèi)物質(zhì)的異常代謝,WVBF主要通過(guò)干擾CFA引起的內(nèi)質(zhì)網(wǎng)應(yīng)激反應(yīng)及線粒體功能紊亂過(guò)程,提高細(xì)胞存活率,使心肌恢復(fù)正常功能.結(jié)論:黃秦艽可以通過(guò)多條信號(hào)通路緩解雪上一枝蒿所致的小鼠心肌毒性作用.
[Abstract]:Aim: to investigate the effect of Gentiana macrophylla Gentiana on the expression of acute toxic genes in heart of mice induced by Artemisia mongolicum by microarray technique and its molecular mechanism. Methods: Twenty-five male Kunming mice were randomly divided into normal control group, (CFA) group and Gentiana macrophylla water extract group (WVBF),). The differentially expressed genes in each group were studied by bioinformatics. Results: WVBF had obvious detoxification effect on acute cardiotoxicity induced by CFA. The function of differentially expressed genes is mainly related to the abnormal metabolism of substances in the body. WVBF improves the survival rate of cells by interfering with the stress response of endoplasmic reticulum and mitochondrial dysfunction induced by CFA, and restores the normal function of myocardium. Conclusion: Gentiana macrophylla can attenuate the myocardial toxicity induced by Artemisia mongolica through several signal pathways.
