不同茶樹品種PPO基因單核苷酸多態(tài)性分析

【摘要】:多酚氧化酶是茶葉加工過程中品質(zhì)形成的關(guān)鍵酶類,在任何一種茶葉加工過程中都起著至關(guān)重要的作用。不同的品種在長期的演化過程中形成了豐富的多酚氧化酶單核苷酸多態(tài)性。本研究主要對著名紅茶產(chǎn)區(qū)選育出的宜興群體品種與引進(jìn)的部分優(yōu)良品種多酚氧化酶基因編碼區(qū)進(jìn)行傳統(tǒng)克隆測序,研究其單核苷酸多態(tài)性,分析不同品種間的遺傳距離遠(yuǎn)近,以及利用不同品種之間的獨(dú)特SNP位點(diǎn)進(jìn)行品種鑒別。結(jié)果表明:32個品種共發(fā)現(xiàn)了120個SNP位點(diǎn),其中81個為非同義突變,39個同義突變,核苷酸多態(tài)性指數(shù)pi為0.011 06,基因的單倍型多樣性(HD)為1.0,說明多酚氧化酶基因具有較高的遺傳多樣性。香山早與梅占的遺傳距離最大,為0.02,這個兩個品種親緣關(guān)系最遠(yuǎn),遺傳距離最小的是鴻雁12號與白葉一號、湘波綠、玉蘭、兩兩之間遺傳距離均為0.001,橫巖3號和烏牛早之間遺傳距離也為0.001,親緣關(guān)系最近。利用不同品種間的多酚氧化酶基因SNP位點(diǎn),可以成功鑒別區(qū)分開32個品種,研究結(jié)果將在茶樹遺傳育種、利用遺傳工程手段提高茶葉品質(zhì)中具有重大意義。
[Abstract]:Polyphenol oxidase (PPO) is the key enzyme in the process of tea processing and plays an important role in any tea processing. Polyphenol oxidase single nucleotide polymorphisms were formed in different varieties during long-term evolution. In this study, the traditional cloning and sequencing of polyphenol oxidase gene coding region of Yixing population selected from famous black tea production area and some imported fine varieties were carried out, the single nucleotide polymorphism was studied, and the genetic distance between different varieties was analyzed. And the unique SNP loci among different varieties were used to identify the varieties. The results showed that there were 120 SNP loci in 32 cultivars, among which 81 were non-synonymous mutations and 39 were synonymous mutations. The nucleotide polymorphism index (pi) was 0.011 and the haplotype diversity (HD) of the gene was 1.0, which indicated that the polyphenol oxidase gene had high genetic diversity. The genetic distance between Xiangshan Zaozao and Meizhan was the largest (0.02). The genetic distance between the two cultivars was the furthest, and the genetic distance was the least between Hongyan 12 and Baiye No. 1, Xiangbo Green, Magnolia. The genetic distance between the two was 0.001, and the genetic distance between Yokyan No. 3 and Wuniuzao was 0.001. The genetic relationship was the most recent. Using the SNP locus of polyphenol oxidase gene among different varieties, 32 varieties can be distinguished successfully. The results will be of great significance in the genetic breeding of tea plants and the improvement of tea quality by means of genetic engineering.
