【摘要】：為了解析甘蔗線條花葉病毒Sugarcane streak mosaic virus(SCSMV)不同分離物HC-Pro基因的分子變異規(guī)律,本研究利用RT-PCR法擴增獲得SCSMV HC-Pro基因的序列,通過生物信息學分析,分別從重組、系統(tǒng)發(fā)生、選擇壓力等方面研究SCSMV HC-Pro基因的分子變異特征。共測定了44條SCSMV HC-Pro基因序列,相似性最低值為70%;HC-Pro基因重組頻率較低,僅發(fā)現(xiàn)3個重組位點,其中一個系首次報道;與先前報道相比,部分新測定云南蔗區(qū)的SCSMV分離物在HC-Pro基因上形成一個新組-第Ⅲ組;HC-Pro基因處于很強的負選擇壓力作用,未發(fā)現(xiàn)正向選擇作用位點。本研究結(jié)果進一步證明SCSMV HC-Pro基因具有高度的遺傳多樣性。
[Abstract]:In order to analyze the rule of molecular variation of HC-Pro gene of sugarcane line mosaic virus (Sugarcane streak mosaic virus (SCSMV), the sequence of SCSMV HC-Pro gene was amplified by RT-PCR method. The sequence of SCSMV HC-Pro gene was sequenced from recombination and phylogeny by bioinformatics analysis. The molecular variation characteristics of SCSMV HC-Pro gene were studied in the aspect of selection pressure. A total of 44 SCSMV HC-Pro gene sequences were sequenced. The lowest similarity was that the recombination frequency of HC-Pro gene was low, and only three recombination sites were found, one of which was reported for the first time. Part of the newly determined SCSMV isolates from sugarcane region of Yunnan Province formed a new group on the HC-Pro gene. The HC-Pro gene of group 鈪，
