【摘要】：目的檢測Beclin1和LC3在卵巢漿液性癌(ovarian serous carcinoma,OSC)中的表達,探討兩者與OSC臨床病理特征及預后的關(guān)系。方法采用免疫組化Max Vision兩步法檢測63例OSC和20卵巢漿液性囊腺瘤組織中Beclin1和LC3蛋白的表達,分析兩者與OSC臨床病理特征及預后的關(guān)系。應用RT-PCR和Western blot法檢測10例新鮮OSC組織和相應癌旁組織中Beclin1和LC3 mRNA和蛋白的表達。結(jié)果在OSC組織中Beclin1和LC3蛋白陽性率(36.51%、33.33%)均低于卵巢漿液性囊腺瘤組織(65.00%、60.00%),差異有統(tǒng)計學意義(P=0.025,P=0.034)。Beclin1蛋白表達與臨床FIGO分期和病理分級有關(guān)(P=0.001,P=0.001),與患者年齡、發(fā)病部位、腫瘤大小及有無盆腔淋巴結(jié)轉(zhuǎn)移無關(guān)(P0.05)。LC3蛋白表達與臨床FIGO分期和盆腔淋巴結(jié)轉(zhuǎn)移有關(guān)(P=0.013,P=0.041),與患者年齡、發(fā)病部位、腫瘤大小和病理分級均無關(guān)(P0.05)。Beclin1和LC3在OSC中的表達呈顯著正相關(guān)(r_s=0.373,P=0.03)。10例新鮮OSC組織中Beclin1和LC3 mRNA的相對表達量(0.581±0.091、0.650±0.090)低于癌旁組織,兩者比較差異有統(tǒng)計學意義(t=8.083,t=6.614;P=0.016,P=0.022)。新鮮OSC組織中Beclin1和LC3蛋白的相對表達量明顯低于癌旁組織,兩者比較差異有統(tǒng)計學意義(P0.05)。Kaplan-Meier生存分析表明,Beclin1和LC3表達與OSC患者預后相關(guān)(P=0.028 3,P=0.018 5)。結(jié)論自噬相關(guān)基因Beclin1和LC3在OSC組織中的表達下降,導致自噬功能減弱,可能與OSC的發(fā)生、發(fā)展和預后不良相關(guān)。
[Abstract]:Objective to detect the expression of Beclin1 and LC3 in ovarian serous carcinoma (ovarian serous carcinoma,OSC) and to explore the relationship between them and the clinicopathological features and prognosis of OSC. Methods the expression of Beclin1 and LC3 in 63 cases of OSC and 20 ovarian serous cystadenomas were detected by immunohistochemical Max Vision two-step method. The relationship between the expression of Beclin1 and LC3 and the clinicopathological features and prognosis of OSC was analyzed. RT-PCR and Western blot were used to detect the expression of Beclin1, LC3 mRNA and protein in 10 fresh OSC tissues and corresponding adjacent tissues. Results the positive rate of Beclin1 and LC3 protein in OSC tissues (36.51%) was lower than that in ovarian serous cystadenoma tissues (65.00%). The difference was statistically significant (P0. 025%, P0. 034). Beclin1 protein expression was correlated with clinical FIGO stage and pathological grade (P0. 001). There was no correlation between tumor size and pelvic lymph node metastasis (P0.05). LC3 protein expression was correlated with clinical FIGO stage and pelvic lymph node metastasis (P0. 013). There was no significant correlation between tumor size and pathological grade (P0.05). The expression of Beclin1 and LC3 in OSC was positively correlated (RSP: 0.373 P0. 03). The relative expression of Beclin1 and LC3 mRNA in 10 fresh OSC tissues (0.581 鹵0.091 鹵0.090) was significantly lower than that in para-cancerous tissues (t8.083t6.614P ~ (0.016) P0. 022). The relative expression of Beclin1 and LC3 protein in fresh OSC tissues was significantly lower than that in paracancerous tissues (P0.05). Kaplan-Meier survival analysis showed that the expression of Beclin1 and LC3 was correlated with the prognosis of OSC patients (P0.028 3, P0. 018 5). Conclusion the decreased expression of autophagy related genes Beclin1 and LC3 in OSC may lead to the decrease of autophagy function, which may be related to the occurrence, development and poor prognosis of OSC.
