尖毛蟲屬Actin Ⅰ、α-TBP和DNA pol α亂序基因的模式研究

【摘要】:研究旨在對尖毛蟲屬內(nèi)現(xiàn)有物種的3種亂序小核基因結(jié)構(gòu)進(jìn)行比較,探討其亂序模式。于湛江湖光紅樹林水域中采集到一個(gè)尖毛蟲屬物種Oxytricha sp.(ZJ),成功擴(kuò)增了其肌動(dòng)蛋白Ⅰ(ActinⅠ)、端粒結(jié)合蛋白(α-TBP)、DNA聚合酶α(DNA polα)3個(gè)亂序基因的完整大核基因序列和完整/部分小核基因序列,并結(jié)合已有資料對比研究了尖毛蟲屬這3個(gè)亂序基因的進(jìn)化。結(jié)果表明:(1)Oxytricha sp.(ZJ)與O.nova的小核ActinⅠ基因具有相同的亂序模式,區(qū)別于其余的尖毛蟲屬物種;在增加尖毛蟲屬物種的基礎(chǔ)上,對前人推測提出了質(zhì)疑,我們認(rèn)為MDS-IES接合處移動(dòng)現(xiàn)象在亂序MDSs之間并非比非亂序MDSs之間更保守。(2)Oxytricha sp.(ZJ)與O.nova的小核α-TBP基因具有相同亂序模式和相似長度的IESs。(3)Oxytricha sp.(ZJ)的小核DNA polα基因亂序模式,區(qū)別于任一已報(bào)道物種,與屬內(nèi)O.trifallax最為相近;谛蛄蟹治,在DNA polα基因中發(fā)現(xiàn)了一例IES轉(zhuǎn)換為MDS的痕跡,以及由此導(dǎo)致原先MDS的丟失。研究發(fā)現(xiàn)在編碼區(qū)內(nèi)IES向MDS的轉(zhuǎn)變,使得本應(yīng)刪除的序列成為基因組永久保留的一部分。
