珠江口3種鯨豚的MHC-I基因多態(tài)性研究

【摘要】:旨在了解珠江口棲息的中華白海豚(Sousa chinensis)、寬脊江豚(Neophocaena phocaenoides)和點(diǎn)斑原海豚(Stenella attenuate)免疫相關(guān)基因MHC-I(Major Histocompatibility Complex)的多態(tài)性及其表達(dá)情況,以期為這3種鯨豚的保育工作提供基礎(chǔ)資料。通過(guò)克隆測(cè)序的方法,首次證實(shí)MHC-I基因在這3種鯨豚體內(nèi)表達(dá)。選擇壓力分析,表明MHC-I基因在這3種鯨豚中均受到強(qiáng)烈的正選擇作用,提示其具有重要的免疫功能。3種鯨豚的MHC-I基因在系統(tǒng)發(fā)育樹(shù)上相互混雜在一起,表明MHC-I基因存在跨物種多態(tài)性。結(jié)合選擇壓力分析和跨物種多態(tài)性,發(fā)現(xiàn)MHC-I基因受到平衡選擇作用。珠江口3種鯨豚MHC-I基因多態(tài)性可能由平衡選擇維持。
[Abstract]:The aim of this study was to investigate the polymorphisms and expression of the immune-related genes MHC-I (Major Histocompatibility Complex) in the Chinese white dolphins (Sousa chinensis), Neophocaena phocaenoides and Stenella attenuate inhabiting the Pearl River Estuary, and to provide basic information for the conservation of the three species of cetaceans. Sequence analysis showed that MHC-I gene was strongly positive in all three species of cetaceans, suggesting that MHC-I gene had important immune function. Mixed MHC-I genes were found in phylogenetic trees of three species of cetaceans, suggesting that MHC-I gene was polymorphic across species. Combining selection stress analysis and cross-species polymorphism, we found that MHC-I gene was affected by balanced selection. The MHC-I gene polymorphism of three species of whales in the Pearl River Estuary may be maintained by balanced selection.
