基于線粒體COⅠ基因部分序列的緬甸安小葉蟬地理種群遺傳多樣性研究

【摘要】:緬甸安小葉蟬Anaka burmensis是一種取食竹子的害蟲,為了探討該物種不同地理種群的遺傳多樣性,本研究首次基于線粒體COⅠ基因部分序列對我國26個(gè)地理種群共241個(gè)樣本進(jìn)行了研究,選取長度為615 bp的基因序列,并運(yùn)用DNASP、MEGA等分析得出,該片段中有546個(gè)保守位點(diǎn)、69個(gè)變異位點(diǎn)和33個(gè)單倍型。種群的單倍型多樣性指數(shù)為0.845,核苷酸多樣性指數(shù)為0.008 77,基因流為0.598 5,種群間的固定系數(shù)為0.729 15,表明種群間遺傳多樣性水平高、遺傳分化大、基因交流水平較低。中性檢驗(yàn)Tajima's D為-1.658 98,0.10P0.05,Fu's Fs值為-5.787,P0.10。分子變異結(jié)果顯示,該物種遺傳變異主要來自種群間,變異百分率為72.92%,而種群內(nèi)的遺傳變異低,僅為27.08%。研究結(jié)果得出該物種遺傳結(jié)構(gòu),可為今后從事葉蟬類昆蟲的分子生物學(xué)研究及該蟲的防治提供理論基礎(chǔ)資料。
