牦牛KLF2基因功能區(qū)克隆及組織表達分析

【摘要】:旨在克隆牦牛KLF2(Krupple-like transcription factor 2,Kruppel樣轉錄因子2)基因功能區(qū)序列,闡明其組織表達規(guī)律,并揭示基因與肌內脂肪含量的相關性。以麥洼牦牛為試驗動物,利用RT-PCR方法克隆KLF2基因功能區(qū)序列,熒光定量PCR技術檢測該基因在各種組織中的表達模式。結果表明,獲得牦牛KLF2基因序列454bp(GenBank登陸號:KX395178),包括KLFs家族典型的3個鋅指結構域;KLF2mRNA在牦牛的肺臟和脂肪組織中存在高水平表達,極顯著高于其他組織(P0.01);KLF2 mRNA的表達水平與背最長肌IMF(Intramuscular Fat,肌內脂肪)含量呈正相關(P0.01,R=0.885)。結果推測KLF2可能作為牦牛肉質性狀的候選基因。
[Abstract]:The aim of this study was to clone the functional region of yak KLF2 (Krupple-like transcription factor 2 Kruppel like transcription factor 2) gene, to elucidate its tissue expression and to reveal the correlation between gene and intramuscular fat content. The functional region of KLF2 gene was cloned from Maiwa yak by RT-PCR method and the expression pattern of the gene in various tissues was detected by fluorescence quantitative PCR. The results showed that 454bp (GenBank accession number: KX395178) of yak KLF2 gene, including three zinc-finger domains of KLFs family, was highly expressed in the lung and adipose tissue of yaks. The expression level of KLF2 mRNA was significantly higher than that of other tissues (P0.01). There was a positive correlation between the expression of KLF2 mRNA and the content of intramuscular fat (IMF (Intramuscular Fat,) in the longissimus dorsi muscle (P0.01). Results it was speculated that KLF2 might be a candidate gene for quality traits of yak.
