電針干預(yù)對(duì)嗎啡耐受形成中小鼠的生物節(jié)律及其相關(guān)鐘基因Per 1和Per 2表達(dá)的影響

【摘要】:目的觀察電針干預(yù)對(duì)嗎啡受形成中小鼠的生物節(jié)律及其相關(guān)鐘基因Per1和Per2表達(dá)的影響。方法將小鼠24只,隨機(jī)分為空白對(duì)照組(n=8),耐受模型組(n=8),電針干預(yù)組(n=8)。耐受模型組、電針干預(yù)組皮下注射鹽酸嗎啡注射液以建立嗎啡耐受模型。電針干預(yù)組每日上午注射嗎啡后開(kāi)始用電針行雙側(cè)"腎俞"穴(BL 23)電針治療。各組在第1、3、5、7d行小鼠光熱甩尾潛伏期測(cè)定。第7 d在測(cè)定痛閾后處死小鼠取視交叉上核(Suprachiasmatic nucleus,SCN)組織,用熒光定量PCR(RT-PCR)方法檢測(cè)小鼠SCN中Per1、Per2基因的表達(dá)變化。結(jié)果 (1)痛閾:耐受模型組和電針干預(yù)組在第1、3、5d注射嗎啡后,測(cè)得痛閾值明顯升高,與其初痛閾相比差異明顯;電針干預(yù)組第7d痛閾與初痛閾相比有差異。(2)自發(fā)性晝夜活動(dòng)節(jié)律:耐受模型組喝電針干預(yù)組與空白對(duì)照組相比,小鼠周期、總活動(dòng)期、振幅結(jié)果均有差異:空白對(duì)照組與耐受模型組和電針干預(yù)組相比小鼠實(shí)驗(yàn)前后振幅差值有差異,耐受模型組與電針治療組相比具有差異。(3)Per1、Per2基因在SCN中的表達(dá)水平:與空白對(duì)照組相比,耐受模型組的相對(duì)表達(dá)量下調(diào);電針干預(yù)組與耐受模型組相比較結(jié)果又明顯差異。結(jié)論電針干預(yù)組方法可以通過(guò)上調(diào)嗎啡耐受模型小鼠的節(jié)律相關(guān)基因Per1、Per2的表達(dá)量來(lái)減緩嗎啡耐受小鼠痛閾值的下降趨勢(shì),延緩耐受的形成并糾正振幅降低為表現(xiàn)的生物節(jié)律紊亂。
[Abstract]:Objective to observe the effects of electroacupuncture intervention on the biological rhythm and related clock gene Per1 and Per2 expression in morphine-induced mice. Methods Twenty-four mice were randomly divided into control group (n = 8), tolerance model group (n = 8) and electroacupuncture intervention group (n = 8). Tolerance model group, electroacupuncture intervention group, subcutaneous injection of morphine hydrochloride injection to establish morphine tolerance model. The intervention group was treated with bilateral "Shenshu" point (BL 23) every morning after morphine injection. The latent period of photothermal tail flick was measured on the 7th day of the 1st day. After the pain threshold was measured, the tissues of superior optic chiasma (Suprachiasmatic nucleus,SCN) were taken and the expression of Per1,Per2 gene in SCN was detected by fluorescence quantitative PCR (RT-PCR). Results (1) pain threshold: the pain threshold of tolerance model group and electroacupuncture intervention group was significantly higher than that of initial pain threshold after 5 days of morphine injection. The pain threshold of electroacupuncture intervention group was different from that of initial pain threshold on the 7th day. (2) spontaneous circadian activity rhythm: compared with the blank control group, the mice cycle and total activity period in the model group were higher than those in the control group. There were differences in amplitude results between blank control group, tolerance model group and electroacupuncture intervention group before and after the experiment. (3) the expression level of Per1,Per2 gene in SCN: compared with the blank control group, the relative expression of Per1,Per2 gene in the tolerance model group was down-regulated, and the results were significantly different between the electroacupuncture intervention group and the tolerance model group. Conclusion the electroacupuncture intervention group can decrease the pain threshold of morphine tolerance mice by up-regulating the expression of Rhythm related gene Per1,Per2 in morphine tolerant mice. Delay the formation of tolerance and correct the biological rhythmic disorder in which the amplitude is reduced to performance.
