橡膠樹WRKY家族轉(zhuǎn)錄因子HbWRKY75基因的克隆及表達(dá)分析

【摘要】:從巴西橡膠樹無性系PR107的葉片組織中克隆了1個(gè)新的WRKY家族轉(zhuǎn)錄因子基因,命名為HbWRKY75,其ORF序列長為603 bp,編碼201個(gè)氨基酸。生物信息學(xué)分析表明,HbWRKY75在N端第110~160氨基酸之間含有1個(gè)典型的WRKY結(jié)構(gòu)域,屬Ⅱ類WRKY轉(zhuǎn)錄因子家族成員,與葡萄、大豆、油棕、陸地棉和擬南芥等同類基因序列的同源性分別為92%、85%、71%、63%和57%。實(shí)時(shí)熒光定量RT-PCR分析表明,HbWRKY75基因在橡膠樹葉片組織中的表達(dá)量最高,而在葉柄組織中的表達(dá)量最低;HbWRKY75基因在橡膠樹衰老晚期葉片中的表達(dá)量高于其他葉片發(fā)育時(shí)期;乙烯利、茉莉酸甲酯(MeJA)、聚乙二醇(PEG)、H_2O_2、NaCl、低溫處理及機(jī)械損傷均可不同程度地誘導(dǎo)HbWRKY75基因的上調(diào)表達(dá);但ABA處理則誘導(dǎo)HbWRKY75基因的下調(diào)表達(dá)。HbWRKY75可能是與橡膠樹葉片衰老和響應(yīng)乙烯等激素刺激相關(guān)的轉(zhuǎn)錄因子,并在橡膠樹響應(yīng)NaCl和低溫等逆境脅迫中可能發(fā)揮調(diào)控作用。
