【摘要】：為評(píng)價(jià)羊白介素2(IL-2)基因和羊口瘡病毒(OrfV)B2L基因真核表達(dá)重組質(zhì)粒聯(lián)合接種小鼠的免疫效果,本研究將pVAX1-B2L、pVAX1-B2L+pVAX1-IL-2及空載體pVAX1和PBS對(duì)照組通過(guò)肌肉注射的方式免疫KM小鼠,并采用ELISA方法和MTT方法檢測(cè)免疫小鼠的體液和細(xì)胞免疫反應(yīng)。結(jié)果表明,pVAX1-B2L+pVAX1-IL-2聯(lián)合免疫組小鼠血清抗體效價(jià)和IL-2水平均顯著高于pVAX1-B2L免疫組,與pVAX1和PBS對(duì)照組相比差異極顯著(p0.01)。而且pVAX1-B2L+pVAX1-IL-2免疫組的脾淋巴細(xì)胞增殖水平高于pVAX1-B2L免疫組、空載體pVAX1組和PBS對(duì)照組(p0.01)。因此,pVAX1-B2L+pVAX1-IL-2聯(lián)合免疫KM小鼠可以誘導(dǎo)特異性體液免疫和細(xì)胞免疫,并且IL-2具有免疫佐劑的作用,為進(jìn)一步將其用于Orf的防制奠定了基礎(chǔ)。
[Abstract]:In order to evaluate the immune effect of recombinant eukaryotic expression plasmid of sheep interleukin 2 (IL-2) gene and sheep mouth sore virus (OrfV) B 2L) gene in mice, pVAX1-B2L,pVAX1-B2L pVAX1-IL-2 and empty vector pVAX1 and PBS control group were injected intramuscularly into KM mice. The humoral and cellular immune responses of immunized mice were detected by ELISA and MTT methods. The results showed that the serum antibody titers and IL-2 levels of mice immunized with pVAX1-B2L pVAX1-IL-2 were significantly higher than those of pVAX1-B2L immunized mice, and the differences were significant compared with those of pVAX1 and PBS control groups (p0.01). The proliferation level of splenic lymphocytes in pVAX1-B2L pVAX1-IL-2 immunized group was higher than that in pVAX1-B2L immunized group, empty vector pVAX1 group and PBS control group (p0.01). Therefore, combined immunization with pVAX1-B2L pVAX1-IL-2 can induce specific humoral immunity and cellular immunity in KM mice, and IL-2 has the function of immune adjuvant, which lays a foundation for further application of pVAX1-B2L pVAX1-IL-2 in the prevention and control of Orf.
