豬戊型肝炎病毒吉林地區(qū)分離株全基因序列的分析

【摘要】:為了了解吉林地區(qū)HEV分子背景和流行特征,對2013年采自吉林地區(qū)養(yǎng)殖場的毒株編號為Ch-S-1的Ⅳ型戊型肝炎病毒(HEV)的全基因序列進行了分析。通過提取病毒RNA,采用RT-PCR方法分段擴增HEV全長基因,兩端采用RACE法擴增,經(jīng)過序列測定和拼接,利用ClusterX1.81、MEGA3.0、Phylop win 3.0軟件分析基因序列。最終得到了HEV Ch-S-1全長基因組(GenBank登錄號EF077630,7 261bp),其中ORF1、ORF2和ORF3分別編碼1 706,674,114個氨基酸。與人源及豬源各基因型HEV相比,Ch-S-1與基因Ⅰ~Ⅲ型的ORF1片段同源性在78.8%~86.9%之間,而與基因Ⅳ型的ORF1片段同源性在93.7%~97.6%。Ch-S-1與基因Ⅰ~Ⅲ型的ORF2片段同源性在87.5%~92.4%之間,而與基因Ⅳ型的ORF2片段同源性在95.9~98.2%。Ch-S-1與基因Ⅰ~Ⅲ型的ORF3片段同源性在75.2%~84.0%之間,而與基因Ⅳ型的ORF3片段同源性在97.5%~99.2%。結果表明:HEV Ch-S-1全序列的基因結構特征與HEVⅣ型基本一致。
[Abstract]:In order to understand the molecular background and epidemic characteristics of HEV in Jilin region, the whole gene sequence of hepatitis IV hepatitis E virus (HEV), named Ch-S-1, which was collected from Jilin breeding farm in 2013, was analyzed. The full-length HEV gene was amplified by RT-PCR method, and the two ends were amplified by RACE method. After sequencing and splicing, the gene sequence was analyzed by cluster X1.81MEGA3.0 win 3.0 software. Finally, the full-length HEV Ch-S-1 genome (GenBank accession number EF077630h7 261bp) was obtained, in which ORF1 ORF2 and ORF3 encoded 1 706674114 amino acids, respectively.
