【摘要】：L-肉堿在脂肪代謝中起重要的作用,本文構(gòu)建了T7啟動子控制下的過表達(dá)質(zhì)粒p ET28a-cai BCD和p ACYCD-cai F-cai T,并將其轉(zhuǎn)入大腸桿菌BL21(DE3)(簡寫DE3)中,得到基因工程菌BL21(DE3)/cai BCD+cai F+cai T(簡寫DE3/BCD+F+T)。將此菌株接種到生長培養(yǎng)基中,并用終濃度為1 mmol/L的IPTG(異丙基硫代半乳糖苷)誘導(dǎo)培養(yǎng),然后收集菌體細(xì)胞加入到含有巴豆甜菜堿鹽酸鹽的轉(zhuǎn)化液中進(jìn)行轉(zhuǎn)化。結(jié)果顯示,IPTG誘導(dǎo)4 h比誘導(dǎo)10 h后轉(zhuǎn)化得到的L-肉堿量要高,轉(zhuǎn)入了過表達(dá)質(zhì)粒的工程菌DE3/BCD+F+T(7.244 g/L)比野生型DE3(0.5 g/L)提高了15倍。該巴豆甜菜堿鹽酸鹽轉(zhuǎn)化生成L-肉堿的反應(yīng)在2 h后基本達(dá)到穩(wěn)定。
[Abstract]:L- carnitine plays an important role in fat metabolism. The overexpression plasmids p ET28a-cai BCD and p ACYCD-cai F-cai T under the control of T7 promoter were constructed and transferred into Escherichia coli BL21 (DE3) (DE3) to obtain genetic engineering bacteria BL21 (DE3) / cai BCD cai F cai T (DE3/BCD F T). The strain was inoculated into the growth medium and cultured with 1 mmol/L IPTG (isopropylthiogalactoside). Then the somatic cells were collected and added to the transformation solution containing crotonine hydrochloride for transformation. The results showed that the amount of L-carnitine was higher after 4 h induction than that after 10 h induction. DE3/BCD F T (7.244 g / L, which was transferred into the overexpression plasmid, was 15 times higher than that of wild type DE3 (0.5 g / L). The conversion of crotonine hydrochloride to L-carnitine was stable after 2 h.
