基于PCR Array技術(shù)探討健脾祛痰化瘀方對脾虛痰濁動(dòng)脈粥樣硬化巴馬小型豬主動(dòng)脈AS信號(hào)通路相關(guān)基因表達(dá)的影響

【摘要】:目的:利用PCR Array技術(shù)觀察健脾祛痰化瘀中藥對脾虛痰濁動(dòng)脈粥樣硬化(AS)巴馬小型豬主動(dòng)脈AS信號(hào)通路相關(guān)基因表達(dá)變化的影響,探討健脾祛痰化瘀方防治AS的可能機(jī)制。方法:將15只巴馬小型豬隨機(jī)分為正常組、模型組和治療組,每組5只,正常組普通飼料喂養(yǎng),模型與治療組給予高熱量高脂飼料喂養(yǎng)并進(jìn)行冠脈球囊擠壓伴拉傷及跑步措施進(jìn)行干預(yù)至24周,制備巴馬小型豬病癥結(jié)合脾虛痰濁AS模型,治療組除上述造模措施外給予健脾祛痰化瘀方灌胃24周。呼吸機(jī)麻醉后取材,提取各組主動(dòng)脈總RNA,應(yīng)用AS信號(hào)通路PCR芯片檢測健脾祛痰化瘀方對脾虛痰濁AS巴馬小型豬主動(dòng)脈AS信號(hào)通路相關(guān)基因mRNA表達(dá)變化的影響。結(jié)果:PCR Array結(jié)果顯示:與空白組相比,模型組顯著上調(diào)的基因共13個(gè),如ACE、BAX、SELE等基因,顯著下調(diào)的基因共16個(gè),如BCL2A1、LDLR、LPL等基因;與模型組相比,治療組顯著上調(diào)的基因共14個(gè),如BCL2A1、LDLR、BCL2等,顯著下調(diào)的基因共13個(gè)如BAX、PTGS1、SOD1等。結(jié)論:健脾祛痰化瘀方可能參與調(diào)控主動(dòng)脈AS信號(hào)通路中脂質(zhì)代謝與凋亡等相關(guān)基因表達(dá)從而起到防治脾虛痰濁AS巴馬小型豬AS形成及穩(wěn)點(diǎn)斑塊的作用。
[Abstract]:Objective: to observe the effect of traditional Chinese medicine for invigorating spleen and removing phlegm and removing blood stasis on the expression of gene related to as signal pathway in aorta of Bama miniature pigs with spleen deficiency and phlegm turbid atherosclerosis (as), and to explore the possible mechanism of Jianpi Quphlegm Huayu recipe in preventing and treating as. Methods: fifteen Bama miniature pigs were randomly divided into normal group, model group and treatment group with 5 in each group. The model and treatment group were fed with high-calorie and high-fat diet and were treated with coronary balloon extrusion with tension and running for 24 weeks to establish the model of as with spleen deficiency and phlegm turbidity in Bama mini-pigs. The treatment group was treated with Jianpi, phlegm and blood stasis decoction for 24 weeks. After ventilator anesthesia, the aorta total RNAs were extracted, and the effects of Jianpi Quphan Huayu recipe on the mRNA expression of as signal pathway related gene in as mini-pig aorta were detected by as signal pathway PCR chip. Results compared with the control group, 13 genes were significantly up-regulated in the model group and 16 genes were down-regulated, such as BCL2A1, LDLRLPL, and 14 genes were significantly up-regulated in the treatment group compared with the control group. For example, BCL2A1, LDLRN, BCL2 and so on, 13 genes were significantly down-regulated, such as BAXA PTGS1, SOD1 and so on. Conclusion: Jianpi Quphan Huayu recipe may be involved in regulating the expression of related genes such as lipid metabolism and apoptosis in aortic atherosclerosis signaling pathway and thus play a role in preventing and treating as with spleen deficiency and phlegm turbidity and stabilizing plaque.
