FASN基因的多態(tài)性及其與中國荷斯坦奶牛產(chǎn)奶性狀的相關(guān)性分析

【摘要】:為了分析脂肪酸合成酶(fatty acid synthase,FASN)基因的多態(tài)性及其與中國荷斯坦奶牛產(chǎn)奶性狀的相關(guān)性,試驗(yàn)選取230頭荷斯坦奶牛,采用限制性片段長度多態(tài)性聚合酶鏈反應(yīng)(Restriction fragment length polymorphism-polymerase chain reaction,RFLP-PCR),對FASN基因3'-UTR區(qū)和外顯子部分片段的多態(tài)性進(jìn)行檢測,并對突變位點(diǎn)與奶牛產(chǎn)奶性狀的相關(guān)性進(jìn)行分析。結(jié)果表明:在所分析的片段中,檢測到3'-UTR區(qū)存在一個(gè)C→T堿基的置換突變(g.51403203CT);該突變位點(diǎn)為Bam HⅠ酶的自然酶切位點(diǎn),酶切后存在CC、TC、TT三種基因型,TT基因型的乳脂率和脂蛋白比顯著高于TC基因型、CC基因型(P0.05),而三種基因型間乳蛋白率和305 d產(chǎn)奶量差異不顯著(P0.05)。
[Abstract]:In order to analyze the polymorphism of fatty acid synthase (fatty acid synthase) gene and its association with milk production traits in Chinese Holstein cows, 230 Holstein cows were selected. Restriction fragment length polymorphism-polymerase chain polymorphism polymerase chain reaction (RFLP-PCR) was used to detect the polymorphism of FASN gene in 3r-UTR region and exon, and the correlation between mutation site and dairy cow milk yield traits was analyzed.
