【摘要】:采用染色體步移技術(shù),從苦蕎(Fagopyrum tataricum Gaertn.)中克隆獲得Ft CHS1基因5'端側(cè)翼序列,共1038 bp,將其命名為PFt CHS1。生物信息學(xué)分析表明,PFt CHS1中A/T堿基含量為63.5%,含有4個(gè)可能的轉(zhuǎn)錄起始位點(diǎn),分別位于起始密碼子上游-684~-734、-692~-742、-920~-970、-929~-979 bp處,該序列包含TATA-Box和CAAT-Box等啟動(dòng)子核心元件以及與光、低溫和激素應(yīng)答等相關(guān)的功能元件。本研究進(jìn)一步構(gòu)建了PFt CHS1-p BI101植物表達(dá)載體,并瞬時(shí)轉(zhuǎn)化擬南芥(Arabidopsis thaliana L.)葉片,結(jié)果顯示該序列可驅(qū)動(dòng)GUS報(bào)告基因的表達(dá)。低溫(4℃)和光照(UV-B)處理苦蕎幼芽后,采用熒光定量PCR技術(shù)分析Ft CHS1基因的表達(dá)量,結(jié)果表明PFt CHS1可響應(yīng)低溫和紫外環(huán)境脅迫,從而引起Ft CHS1基因表達(dá)量發(fā)生變化。
[Abstract]:The chromosome step technique was used to study the genetic properties of Tartary buckwheat (Fagopyrum tataricum Gaertn.)
