高光脅迫下壇紫菜定量PCR內(nèi)參基因的篩選

【摘要】:為了篩選不同高光光強(qiáng)脅迫下壇紫菜中表達(dá)水平最為穩(wěn)定的內(nèi)參基因,本研究采用qRT-PCR技術(shù)測(cè)定了植物中常用的6種內(nèi)參基因UBC、TUB、18S、EF2、ACT和GAPDH在不同光照強(qiáng)度下培養(yǎng)的壇紫菜藻體中的絕對(duì)表達(dá)水平,并采用比較Ct值法、Ge Norm、Bestkeeper和NormFinder軟件分析4種方法對(duì)6種候選內(nèi)參基因的表達(dá)穩(wěn)定性進(jìn)行評(píng)估。結(jié)果顯示,UBC的Ct值變化范圍最小,18S的Ct值變化范圍最大;ge Norm軟件分析結(jié)果顯示最穩(wěn)定的內(nèi)參基因?yàn)閁BC和EF2;Bestkeeper和NormFinder軟件分析結(jié)果類似,UBC基因的穩(wěn)定值M均為最低值(分別為0.044和0.80),說(shuō)明其表達(dá)水平最穩(wěn)定。研究表明,UBC基因可以作為不同光照條件下壇紫菜基因表達(dá)qRT-PCR分析的最適內(nèi)參基因。
[Abstract]:In order to screen the most stable internal reference gene in porphyra sinensis under different high light intensity stress, In this study, qRT-PCR technique was used to determine the absolute expression levels of six common internal reference genes UBCU, TUBX, TUBN, EF-2ACT and GAPDH in different light intensity cultures of porphyra chinensis. Four methods were used to evaluate the expression stability of six candidate internal reference genes by comparing Ct value method with GE NormBestkeeper and Norm Finder software. The results showed that the range of Ct value of UBC was the smallest and the change range of Ct value of 18s was the largest. The result of software analysis showed that the most stable internal reference group was the most stable because UBC and EF2Bestkeeper and NormFinder software analysis results were similar to the stability value M of UBC gene. It was 0.044 and 0.80, respectively, indicating that its expression level was the most stable. The results showed that UBC gene could be used as the most suitable internal reference gene for qRT-PCR analysis of porphyra chinensis gene expression under different illumination conditions.
