人食管鱗狀細(xì)胞癌中微RNA-1250及其宿主基因AATK的表達(dá)及甲基化狀態(tài)分析

【摘要】:目的:探討微RNA-1250(microRNA-1250,miR-1250)及其宿主基因細(xì)胞凋亡關(guān)聯(lián)酪氨酸激酶(apoptosis-associated tyrosine kinase,AATK)在食管鱗狀細(xì)胞癌(esophageal squamous cell carcinoma,ESCC)發(fā)生和發(fā)展中的作用。方法:分別采用實(shí)時(shí)熒光定量PCR法和甲基化特異性PCR法檢測(cè)AATK基因在食管癌細(xì)胞系和ESCC組織及其相應(yīng)癌旁正常組織中的表達(dá)情況和甲基化狀態(tài),同時(shí)采用實(shí)時(shí)熒光定量PCR法檢測(cè)miR-1250在ESCC組織及其相應(yīng)癌旁正常組織中的表達(dá)情況。然后,分析miR-1250表達(dá)和AATK基因表達(dá)及其甲基化狀態(tài)分別與ESCC組織病理學(xué)特征的關(guān)系,以及三者之間的相關(guān)性。結(jié)果:5-雜氮-2’-脫氧胞苷(5-aza-2’-deoxycytidine,5-Aza-dC)處理后,4株食管癌細(xì)胞系中AATK基因表達(dá)水平明顯升高(P值均0.05),并且其甲基化程度明顯降低。在ESCC組織中,AATK mRNA平均表達(dá)量低于癌旁正常組織(P0.05),AATK基因甲基化率明顯高于癌旁正常組織(P0.05);AATK mRNA相對(duì)表達(dá)量和AATK基因甲基化率均與淋巴結(jié)轉(zhuǎn)移、組織分化程度及TNM分期密切相關(guān)(P值均0.05),而與患者年齡和性別無關(guān)(P值均0.05)。同時(shí),ESCC組織中miR-1250相對(duì)表達(dá)量明顯低于癌旁正常組織(P0.05),并與淋巴結(jié)轉(zhuǎn)移、組織分化程度及TNM分期密切相關(guān)(P值均0.05),而與患者年齡和性別無關(guān)(P值均0.05)。ESCC組織中miR-1250與AATK mRNA表達(dá)明顯相關(guān)(P0.05),而且miR-1250和AATK mRNA表達(dá)與AATK基因甲基化狀態(tài)也均明顯相關(guān)(P值均0.05)。結(jié)論:ESCC的發(fā)生和發(fā)展可能與miR-1250和AATK低表達(dá)以及AATK基因高甲基化狀態(tài)有關(guān),而且miR-1250與AATK mRNA表達(dá)具有一致性。
[Abstract]:Aim: to investigate the role of microRNA-1250 (microRNA-1250 miR-1250) and its host gene apoptosis-associated tyrosine kinase (apoptosis-associated tyrosine kinase- AATK) in the pathogenesis and development of esophageal squamous cell carcinoma (esophageal squamous cell carcinomaESCC). Methods: the expression and methylation status of AATK gene in esophageal carcinoma cell line, ESCC and its adjacent normal tissues were detected by real-time fluorescence quantitative PCR and methylation specific PCR, respectively. The expression of miR-1250 in ESCC and its adjacent normal tissues was detected by real-time fluorescent quantitative PCR. Then, the relationship between the expression of miR-1250 and AATK gene and the histopathological characteristics of ESCC was analyzed. Results the expression level of AATK gene in 4 esophageal cancer cell lines treated with 5 aza-2aza-deoxycytidine 5-Aza-dC was significantly increased (P < 0.05), and its methylation degree was significantly decreased. The average expression of AATK mRNA in ESCC was significantly lower than that in adjacent normal tissues (P0.05), and the methylation rate of AATK mRNA and AATK gene methylation were significantly higher than those in adjacent normal tissues (P0.05) and lymph node metastasis. Tissue differentiation and TNM stage were closely related (P 0.05), but not related to age and sex (P 0.05). At the same time, the relative expression of miR-1250 in ESCC was significantly lower than that in adjacent normal tissues (P0.05), and the expression of miR-1250 was associated with lymph node metastasis. Tissue differentiation and TNM stage were closely correlated (P 0.05), but not related to age and sex (P 0.05). MiR-1250 was significantly correlated with AATK mRNA expression in ESCC (P0.05), and miR-1250 and AATK mRNA expression were significantly correlated with AATK gene methylation (P 0.05). Conclusion the occurrence and development of the cell carcinoma may be related to the low expression of miR-1250 and AATK and the hypermethylation of AATK gene, and miR-1250 is consistent with the expression of AATK mRNA.
