【摘要】：目的:檢測(cè)Dicer1在卵巢癌間質(zhì)腫瘤相關(guān)成纖維細(xì)胞中的表達(dá)情況,探討其對(duì)DNA雙鍵斷裂損傷(DSB)修復(fù)相關(guān)基因的影響。方法:應(yīng)用基因集合富集分析(GSEA)方法分析卵巢癌間質(zhì)和正常卵巢間質(zhì)基因表達(dá)公共數(shù)據(jù)庫GSE40595,探索Dicer1對(duì)DSB修復(fù)相關(guān)通路及關(guān)鍵基因的調(diào)控作用,并篩選出相關(guān)差異基因。應(yīng)用重組人轉(zhuǎn)化生長因子β1(h TGFβ1)細(xì)胞因子誘導(dǎo)成纖維細(xì)胞系MRC5細(xì)胞活化成為腫瘤相關(guān)成纖維細(xì)胞(CAFs),即MRC5-CAFs。以靶向Dicer1基因的siRNA轉(zhuǎn)染MRC5-CAFs,下調(diào)其Dicer1表達(dá)水平。qRT-PCR、Western blot法分別檢測(cè)Dicer1及分析所得相關(guān)基因,如XRCC6、TP53BP1、H2AFX、BRCA1、ATR、RAD51 mRNA水平及Dicer1和DSB損傷修復(fù)關(guān)鍵基因γ-H2AX(磷酸化H2AX)、RAD51蛋白水平。熒光共聚焦檢測(cè)細(xì)胞核蛋白γ-H2AX表達(dá);采用CCK8試驗(yàn)檢測(cè)細(xì)胞活性。結(jié)果:GSEA分析結(jié)果顯示,Dicer1表達(dá)水平與DSB(NES=1.7942109,FDRq=0.0067545176)及DNA損傷修復(fù)(NES=2.4433048,FDRq=0)顯著正相關(guān),進(jìn)一步通過相關(guān)分析篩選出在上述通路中與Dicer1表達(dá)正相關(guān)的顯著基因集。沉默MRC5-CAFs細(xì)胞中Dicer1表達(dá)后,DSB相關(guān)基因,如XRCC6、TP53BP1、H2AFX及損傷修復(fù)相關(guān)基因BRCA1、ATR、RAD51均較對(duì)照組顯著下降,與GSEA分析結(jié)果一致。熒光共聚焦顯微鏡檢測(cè)到沉默Dicer1 MRC5-CAFs細(xì)胞內(nèi)聚集較少的DSB。CCK8試驗(yàn)結(jié)果顯示,MRC5-CAFs細(xì)胞在20μmol/L順鉑作用下,Dicer1-si組不同時(shí)間梯度的細(xì)胞活性率均低于NC-si組。結(jié)論:Dicer1在卵巢癌間質(zhì)成纖維細(xì)胞中正向調(diào)控DNA損傷修復(fù)相關(guān)基因,抑制Dicer1表達(dá)后其對(duì)順鉑敏感性顯著增加。
[Abstract]:Aim: to detect the expression of Dicer1 in ovarian cancer stromal tumor-associated fibroblasts and to investigate the effect of Dicer1 on DNA double bond break injury (DSB) repair related genes. Methods: GSE40595, a common database for gene expression in ovarian cancer stromal and normal ovarian stroma, was analyzed by gene set enrichment analysis (GSEA). The regulatory effects of Dicer1 on DSB repair pathway and key genes were explored, and the differentially related genes were screened out. Recombinant human transforming growth factor 尾 1 (hTGF 尾 1) cytokines were used to induce the activation of MRC5 cells into tumor-associated fibroblasts (CAFs), that is, MRC5-CAFs. The Dicer1 gene was transfected into MRC5-CAFs by siRNA targeting Dicer1 gene. The expression level of Dicer1 was down-regulated. QRT-PCRN Western blot assay was used to detect Dicer1 and analyze the related genes, such as the level of XRCC6TP-53BP1AP-H2AFXT1-BRCA1AP-RAD51 mRNA and the RAD51 protein level of 緯 -H2AX (phosphorylated H2AX), a key gene for damage repair of Dicer1 and DSB. The expression of nuclear protein 緯 -H2AX was detected by confocal fluorescence, and the cell activity was detected by CCK8 assay. Results the expression level of Dicer1 was positively correlated with DSB (NES 1.7942109) and DNA damage repair (NESN 2.4433048 FDRq0). The significant gene sets associated with Dicer1 expression in the above pathway were screened by correlation analysis. After the expression of Dicer1 was silenced in MRC5-CAFs cells, the expression of DSB related genes, such as XRCC6, TP53BP1H2AFX, and the damage repair related gene BRCA1 / ATRN RAD51, were significantly lower than those in the control group, which were consistent with the results of GSEA analysis. The results of fluorescence confocal microscopy showed that the DSB.CCK8 assay of silent Dicer1 MRC5-CAFs cells showed that the cell viability of DSB.CCK8 cells in dicer1-si group was lower than that in NC-si group under 20 渭 mol / L cisplatin. Conclusion the gene associated with DNA damage repair is positively regulated by the expression of dicer1 in ovarian cancer stromal fibroblasts, and its sensitivity to cisplatin is significantly increased after the inhibition of Dicer1 expression.
