西伯利亞鱘Sox17基因cDNA全長(zhǎng)的克隆及其表達(dá)分析

【摘要】:采用RACE技術(shù)獲得西伯利亞鱘(Acipenser baerii)Sox17基因cDNA全長(zhǎng),序列分析表明,西伯利亞鱘Sox17基因cDNA全長(zhǎng)2 671 bp,包括588 bp的5'端非翻譯區(qū),901 bp的3'端非翻譯區(qū)和編碼393個(gè)氨基酸殘基的1 183 bp開(kāi)放閱讀框。氨基酸序列比對(duì)顯示,該基因具較高的保守性,與塞內(nèi)加爾多鰭魚(yú)(Polypterus senegalus)的相似度最高為72%。系統(tǒng)進(jìn)化分析表明,西伯利亞鱘與塞內(nèi)加爾多鰭魚(yú)聚為一支,且支持率為95%。以延伸因子efla(elongation factor 1-alpha)作為內(nèi)參基因,對(duì)Sox17基因在西伯利亞鱘各組織的相對(duì)表達(dá)量進(jìn)行分析,發(fā)現(xiàn)Sox17基因在不同組織中均有表達(dá),但表達(dá)具有明顯的組織差異性。在腦中表達(dá)量最高,而在腎、血液、眼、腸和胸鰭5個(gè)組織中表達(dá)量均較低。同時(shí),Sox17基因在西伯利亞鱘不同發(fā)育時(shí)期均有表達(dá),但表達(dá)具有一定的時(shí)間差異性。在寬神經(jīng)板期的表達(dá)量高,相當(dāng)于表達(dá)量最低點(diǎn)11日齡仔魚(yú)的167倍。本研究為量化西伯利亞鱘胚胎發(fā)育分化過(guò)程中相關(guān)基因提供了參照工具,同時(shí)可為今后深入研究西伯利亞鱘Sox17在胚胎發(fā)育過(guò)程中的作用提供基礎(chǔ)參考資料。
[Abstract]:The full-length cDNA of (Acipenser baerii) Sox17 gene of Acipenser Siberian Sturgeon was obtained by race technique. The full-length cDNA of Sox17 gene of Acipenser Siberia Sturgeon is 2 671 BP, including 588 BP 5'untranslated region 901 BP untranslated region and 1 183 BP open reading frame encoding 393 amino acid residues. Amino acid sequence alignment showed that the gene was highly conserved and had the highest similarity to the Senegalese polyfin fish (Polypterus senegalus). Phylogenetic analysis showed that the Siberian Sturgeon and the Senegalese polyfin were one and the support rate was 95. The relative expression of Sox17 gene in various tissues of Acipenser sibirica was analyzed by using the extension factor efla (elongation factor 1-alpha as internal reference gene. It was found that Sox17 gene was expressed in different tissues, but the expression of Sox17 gene was different in different tissues. The highest expression was found in the brain, but low in the kidney, blood, eye, intestine and pectoral fin. At the same time, Sox17 gene was expressed in different developmental stages of Acipenser sibirica, but the expression had some time difference. The expression level in the broad nerve plate stage was 167 times of that of the 11 day old larva at the lowest level of expression. This study provides a reference tool for quantifying the genes involved in embryonic development and differentiation of Acipenser Siberian Sturgeon, and provides a basic reference for the further study of the role of Sox17 in embryonic development of Acipenser Siberian Sturgeon.
