【摘要】：花是被子植物主要的繁殖器官,在繁育后代的過程中,發(fā)揮著極其重要的作用,PISTILLATA(PI)基因作為控制花器官發(fā)育的B類功能基因中的一員,在花器官發(fā)育中起到重要的作用。為探究PI基因在花瓣和雄蕊發(fā)育中的功能,本文以擬南芥(Arabidopsis thaliana)中的PI基因作為研究對象,利用PCR技術(shù)從擬南芥花序c DNA擴(kuò)增出At PI基因,構(gòu)建植物表達(dá)載體(p ROKⅡ-At PI)并進(jìn)行煙草(Nicotiana tobacum)轉(zhuǎn)化。轉(zhuǎn)基因植株的PCR檢測結(jié)果表明,At PI基因已經(jīng)整合到了煙草基因組中。在T2代植株中,通過實(shí)時定量熒光PCR檢測顯示,At PI在m RNA水平也均有表達(dá)。過量表達(dá)PI的轉(zhuǎn)基因煙草在花器官中存在明顯表型,與野生型相比主要表現(xiàn)為轉(zhuǎn)基因植株花冠變小,雄蕊縮短,果實(shí)畸形且子房基部比野生型長5~10 mm,上述結(jié)果表明At PI基因是特異性參與雄蕊和花瓣的發(fā)育并起著至關(guān)重要的作用。
