芥菜型油菜多室基因Bjmc2的精細(xì)定位

【摘要】:芥菜型多室油菜的產(chǎn)量比普通兩室油菜更高,定位乃至克隆多室基因可為油菜遺傳改良及解釋多室角果形成機(jī)制創(chuàng)造條件。本研究通過(guò)驗(yàn)證JD11-2家系衍生群體僅在BjMc2位點(diǎn)上存在差異,可用于BjMc2的定位。采用AFLP結(jié)合BSA法分析BC_5和BC_6群體,篩選到1個(gè)與BjMc2連鎖的AFLP標(biāo)記并轉(zhuǎn)化為SCAR標(biāo)記SC1;谠揂FLP標(biāo)記序列信息,利用白菜同源序列設(shè)計(jì)SSR引物和SCAR引物,獲得11對(duì)SSR標(biāo)記和1對(duì)SCAR標(biāo)記。通過(guò)在芥菜型油菜BAC文庫(kù)中的挑選,獲得2個(gè)覆蓋目標(biāo)區(qū)域的單克隆,由此開(kāi)發(fā)1個(gè)SSR標(biāo)記。將獲得的SCAR和SSR標(biāo)記掃描BC7群體,構(gòu)建了兩室性狀基因BjMc2的遺傳連鎖圖,兩側(cè)最近標(biāo)記ZX17和BACsr96與目標(biāo)基因之間的遺傳距離分別為0.048 cM和0.340 cM,并定位到白菜A7 Scaffold 000019的946~1014 kb之間,約68 kb物理距離。
[Abstract]:The yield of mustard multi-compartment rapeseed was higher than that of common two-compartment rape. Mapping and even cloning of multi-compartment genes could create conditions for genetic improvement and explanation of the formation mechanism of multi-compartment pods in Brassica napus. In this study, it was verified that only BjMc2 loci were different in JD11-2 family derived populations, which could be used to localize BjMc2. An AFLP marker linked to BjMc2 was screened and transformed into scar marker SC1 by AFLP combined with BSA method. Based on the AFLP marker sequence information, SSR primers and scar primers were designed by using the homologous sequence of Chinese cabbage, and 11 pairs of SSR markers and 1 pair of scar markers were obtained. By selecting the BAC library of Brassica napus, two monoclonal clones covering the target region were obtained and an SSR marker was developed. The BC7 population was scanned with scar and SSR markers, and the genetic linkage map of the two-compartment trait gene BjMc2 was constructed. The genetic distances between the two most recent markers ZX17 and BACsr96 and the target gene were 0.048 cm and 0.340 cm, respectively, and the genetic distance was between 9461014 kb and 9461014 kb of Chinese cabbage A7 Scaffold 000019. About 68 kb physical distance.
