三七總皂苷對(duì)腦出血大鼠神經(jīng)細(xì)胞凋亡及相關(guān)基因的影響

【摘要】:目的:研究三七總皂苷對(duì)腦出血大鼠神經(jīng)細(xì)胞凋亡及相關(guān)基因的影響.方法:將大鼠隨機(jī)分為假手術(shù)組、模型組和三七總皂苷組,利用膠原酶注射尾狀核法制作腦出血模型,利用免疫組化染色法及細(xì)胞凋亡染色法檢測(cè)各組大鼠Bcl-2、Bax、Caspase-3及TUNEL陽性細(xì)胞數(shù)目.結(jié)果:在6 h、2 d、3 d、5 d、7 d共5個(gè)時(shí)間點(diǎn),模型組、三七總皂苷組大鼠神經(jīng)細(xì)胞凋亡情況、Bcl-2、Bax、Caspase-3與假手術(shù)組相比差異均顯著,具有統(tǒng)計(jì)學(xué)意義(P0.05);三七總皂苷組大鼠神經(jīng)細(xì)胞凋亡情況、Bcl-2、Bax、Caspase-3與模型組相比差異均顯著,具有統(tǒng)計(jì)學(xué)意義(P0.05).結(jié)論:三七總皂苷治療可以顯著下調(diào)Bax、Caspase-3的表達(dá)水平,提高Bcl-2的表達(dá)水平,減少神經(jīng)細(xì)胞凋亡,降低腦出血后繼發(fā)性損傷.
[Abstract]:Aim: to study the effect of Panax notoginseng saponins on neuronal apoptosis and related genes in rats with intracerebral hemorrhage. Methods: rats were randomly divided into sham-operation group, model group and panax notoginseng saponins group. Intracerebral hemorrhage model was made by injection of collagenase into caudate nucleus. Immunohistochemical staining and apoptosis staining were used to detect the number of Caspase-3 and Tunel positive cells. Results: the apoptosis of nerve cells in model group, panax notoginseng saponins group and sham operation group were significantly different from those in sham operation group at 6 h, 2 d, 3 d, 5 d and 7 d, and the apoptosis of nerve cells in model group and panax notoginseng saponins group were significantly higher than those in sham operation group. The apoptosis of nerve cells in the panax notoginseng saponins group was significantly different from that in the model group. Conclusion: panax notoginseng saponins can significantly down-regulate the expression of Caspase-3, increase the expression of Bcl-2, reduce neuronal apoptosis and reduce the secondary injury after intracerebral hemorrhage.
