【摘要】:本研究通過c DNA末端快速擴(kuò)增法(RACE)首次克隆得到凡納濱對蝦(Litopenaeus vannamei)堿性磷酸酶(alkaline phosphatase,ALP)和酸性磷酸酶(acid phosphatase,ACP)基因的c DNA全長序列,其中ALP基因全長序列1 910 bp,編碼536個氨基酸;ACP基因全長序列1 544 bp,編碼439個氨基酸。鹽度調(diào)控設(shè)3個鹽度組:31(對照)、16和3。養(yǎng)殖45 d后,熒光定量PCR測定對蝦肝胰腺、肌肉、胃、腸和鰓5個組織中的ALP和ACP基因m RNA的表達(dá)情況。RT-PCR顯示,ALP基因在5個組織中普遍表達(dá),在肝胰腺中表達(dá)量最高(P0.05),腸道中的表達(dá)量最低(P0.05);ACP基因在5個組織中也均表達(dá),在肝胰腺和鰓中表達(dá)量明顯高于其他3個組織(P0.05)。鹽度應(yīng)答實(shí)驗(yàn)表明,不同鹽度條件下,不同組織中,ALP和ACP基因存在不同的表達(dá)模式。在胃和鰓組織中,ALP基因在31鹽度組的表達(dá)量顯著高于其他兩組(P0.05);在肝胰腺和腸道中,ALP基因在16鹽度下的表達(dá)量明顯低于其他兩組(P0.05);在肌肉中,ALP基因的表達(dá)與其他組織均不同,31鹽度組表達(dá)量最高,而16鹽度組最低(P0.05);在肝胰腺中,ACP基因在31鹽度組的表達(dá)量明顯低于其他鹽度組(P0.05);在肌肉和鰓兩個組織中,ACP基因在31鹽度下的表達(dá)量明顯高于其余兩個鹽度組(P0.05);在腸道中,ACP基因的表達(dá)量在3鹽度組明顯高于其他兩組(P0.05);而在胃中,ACP基因在3個鹽度下的表達(dá)量沒有顯著性差異。上述結(jié)果為研究低鹽條件下凡納濱對蝦的磷酸酶基因的變化機(jī)制提供了參考。
[Abstract]:In this study, the cDNA sequences of the alkaline phosphatase (ALP) and acid phosphatase (ACP) gene of Litopenaeus vannamei) were cloned for the first time by rapid amplification of cDNAs. The total length of ALP gene was 1 910 BP, the total length of ACP gene encoding 536 amino acids was 1 544 BP, and that of ALP gene was 439 amino acids. The salinity regulation was divided into 3 salinity groups: 1: 31 (control group: 16 and 3. After 45 days of culture, the expression of ALP and ACP mRNA in the liver and pancreas, muscle, stomach, intestine and Gill of prawn was detected by fluorescence quantitative PCR. RT-PCR showed that ALP gene was widely expressed in 5 tissues. The highest expression of P0.05 was found in the hepatopancreas, and the lowest expression of P0.05ACP was found in the intestine. The expression of ACP in the hepatopancreas and gills was significantly higher than that in the other three tissues. The salinity-response experiments showed that there were different expression patterns of ALP and ACP genes in different tissues under different salinity conditions. The expression of ALP gene in gastric and Gill tissues was significantly higher in the 31 salinity group than in the other two groups; in the hepatopancreas and intestine, the expression of the ALP gene in the liver, pancreas and intestine was significantly lower than that in the other two groups at 16 salinity; in the muscle, the ALP gene expression was similar to that in the other two groups. The highest expression level was observed in all tissues with different salinity. The expression of ACP gene in the hepatopancreas was significantly lower than that in the other salinities group, and the expression of ACP gene in the muscle and Gill tissues was significantly higher than that in the other two salinities groups (P0.05A), while the expression of ACP gene in the hepatopancreas was significantly lower than that in the other two salinities groups (P 0.05), while the expression of ACP gene in the muscle and Gill tissues was significantly higher than that in the other two salinity groups. The expression of ACP gene in the intestine was significantly higher in the 3 salinity group than in the other two groups, but there was no significant difference in the expression of ACP gene in the stomach under the three salinity conditions. These results provide a reference for studying the mechanism of phosphatase gene change in Penaeus vannamei under low salt condition.
