馬鈴薯小熱激蛋白基因sHSP-F的克隆及植物表達載體的構(gòu)建

【摘要】:熱激蛋白(heat shock proteins,HSP)是生物體在不利環(huán)境條件因素刺激下應激合成的一組在進化上高度保守的蛋白質(zhì)。前期轉(zhuǎn)錄組測序的結(jié)果發(fā)現(xiàn)馬鈴薯Favorita的小熱激蛋白(small heat shock proteins,s HSPs)基因(PGSC0003DMG400009255)在接種晚疫病菌(Phytophthora infestans)24 h后表達量顯著上調(diào)。因此,以馬鈴薯Favorita為材料,根據(jù)PGSC0003DMG400009255基因序列設計引物,并在引物5'端加上Bam HⅠ和SalⅠ酶切位點,從接種P.infestans 24小時后的Favorita的RNA中通過RT-PCR的方法獲得PGSC0003DMG400009255的基因片段,并命名為s HSP-F,該基因最大開放閱讀框(ORF)為594 bp,編碼197個氨基酸。通過酶切連接將s HSP-F連接至表達載體p CAMBIA1301中。通過測序和酶切驗證,表明s HSP-F基因成功克隆到表達載體中,該工作為進一步研究該基因的功能提供了基礎。
[Abstract]:Heat shock protein (shock) is a group of highly evolutionarily conserved proteins stimulated by adverse environmental conditions. The results of transcriptome sequencing showed that the expression of small heat shock protein (HSPs) gene PGSC0003DMG40000009255 in potato Favorita was significantly up-regulated after inoculation with Phytophthora infestans)24.
