中間錦雞兒CiABR17基因的克隆及表達(dá)分析

【摘要】:該研究采用blastn比對(duì)方法,從中間錦雞兒干旱脅迫抑制消減雜交文庫(kù)篩選出ABA響應(yīng)蛋白17基因(ABR17)的EST序列。RACE擴(kuò)增得到cDNA序列594bp,其中ORF為471bp,編碼157個(gè)氨基酸,預(yù)測(cè)蛋白質(zhì)分子量為16.44kD,等電點(diǎn)為4.93,平均疏水指數(shù)為-0.126,是一種親水蛋白。實(shí)時(shí)熒光定量PCR檢測(cè)發(fā)現(xiàn),中間錦雞兒CiABR17基因在脫水、NaCl和熱脅迫下均有不同程度表達(dá),說(shuō)明CiABR17基因與中間錦雞兒響應(yīng)逆境脅迫有關(guān);過(guò)表達(dá)CiABR17擬南芥株系的鮮重、干重和蓮座直徑均極顯著高于野生型株系。
[Abstract]:In this study, the blastn sequence 594 BP was amplified from drought stress suppression subtractive hybridization library of Caragana Intermediate Caragana by EST sequence. Race amplification of ABR17 gene, in which ORF is 471 BP, encodes 157 amino acids. The predicted molecular weight of protein is 16.44 kD, the isoelectric point is 4.93, and the average hydrophobic index is -0.126, which is a hydrophilic protein. The results of real-time fluorescence quantitative PCR showed that CiABR17 gene was expressed in different degree under dehydration NaCl and heat stress, which indicated that CiABR17 gene was related to stress response of Caragana intermedia, and the fresh weight of Arabidopsis thaliana strain CiABR17 was overexpressed. Both dry weight and rosette diameter were significantly higher than those of wild-type lines.
